摘  要:背景:有研究表明Survivin作为一种凋亡蛋白抑制蛋白,在胚胎植入过程中可能起到抑制子宫内膜凋亡及诱导子宫内膜蜕膜化和促使滋养层细胞入侵的作用,但是目前关于Survivin在胚胎着床方面的报道较少。目的:观察凋亡蛋白抑制因子Survivin在早孕小鼠子宫内膜中的表达及对胚胎着床的作用。设计、时间及地点:随机分组设计,实验于2008-03/06在重庆医科大学医学检验中心完成。材料:6~8周龄清洁级NIH小鼠76只,体质量20~25g;兔抗人Survivin多克隆抗体为美国Santacruz公司产品。方法:①根据阴道涂片结果鉴别雌鼠的动情周期,动情后期雌鼠设为未孕组,记为妊娠第0天。雌雄鼠按2∶1比例自然合笼,次晨7时检查雌鼠阴栓阳性者即为妊娠,定为妊娠第1天,依次计算妊娠天数。②取妊娠4d的小鼠20只随机抽签法分为2组,每组10只。对照组:双侧子宫角均注射5μL正常兔血清IgG;实验组:左侧子宫角注射Survivin1∶80比例抗体5μL,右侧子宫角正常兔血清IgG5μL对照。妊娠第8天剖腹取出子宫,大体观察并统计胚胎着床数。主要观察指标:①免疫组织化学检测Survivin在未孕及妊娠第2,3,4,5,6,7天各组小鼠子宫内膜的表达。②子宫角注入Survivin抗体,统计胚胎着床数。结果:①免疫组织化学检测妊娠组Survivin蛋白的表达明显高于未孕组(P<0.05),且随着妊娠天数的增加,子宫内膜Survivin蛋白的表达逐渐增加,到孕第6天达到高峰,随后下降,但孕第7天仍明显高于未孕组(P<0.05)。②实验组左右侧子宫胚胎着床数分别为1.4000±0.5164和6.4000±1.0750;对照组左右侧子宫胚胎着床数为6.5000±1.0801和6.2000±0.9189;实验组注射抗体的左侧胚胎着床数较右侧明显下降(P<0.05);对照组左右侧子宫胚胎着床数差异无显著性意义(P>0.05)。结论:Survivin在早孕小鼠子宫内膜持续表达,并参与胚胎的着床调控过程。BACKGROUND: Survivin as a inhibitor of apoptosis factor, is maybe involved in apoptosis inhibition of endometrium, coordinating decidualization of endometrium and invasion of trophoblastic cells, however there are few reports on Survivin embryo implantation. OBJECTIVE: To investigate the expression of Survivin in mouse endometrium during early pregnancy and its role in embryo implantation. DESIGN, TIME AND SETTING: The experiment was performed in randomization at Center for Medicine Probation at the Chongqing Medical University between March 2008 and June 2008. MATERIALS: Seventy clean NIH mice aged 6 to 8 weeks weighing 20 to 25 g were enrolled; the rabbit anti-person Survivin multi-clonal antibody was purchased from American Santa the cruz Corporation product. METHODS: ①The female mice estrum was identified based on vaginal smears results, mice of later estrum were designed to be non-pregnancy group, recorded as day 0 of pregnancy. Mice of female and male were housed together at a ratio of 2:1 overnight and the confirmation of pregnancy was determined at 7 o’clock on the second day by female mice’s vaginal plug, the positive result was pregnant, identified as day 1 of pregnancy, then to calculate days of pregnancy as the day followed by. ②Twenty pregnant mice were randomly divided into control and treatment groups with 10 mice in each group. The control group underwent normal rabbit blood serum IgG (5 μL) injection into the bilateral uterine horns; the left uterine horn of each mouse was injected with 1:80 anti-Survivin polyclonal antibodies (5 μL) in the treatment group, while the right uterine horn was injected with normal rabbit blood serum IgG (5 μL). The mice were sacrificed on day 8 of pregnancy and then the number of implantation embryos was recorded with visual observation.
