摘 要:研究了利用2-丙烯基芥子油苷外标法定量芥蓝中芥子油苷的可行性,并对芥蓝中芥子油苷种类及含量进行报道。结果表明,2-丙烯基芥子油苷在14.2U硫酸酯酶的脱硫下,经过高效液相色谱检测,峰面积与浓度呈正比,建立回归方程:y=3E-07x+0.001,相关系数达到0.9994。回收及对比实验表明,2-丙烯基芥子油苷外标法定量的结果准确可信,可以代替昂贵的内标苯甲基芥子油苷。经HPLC/ESI-MS鉴定和外标法定量,在芥蓝菜薹中存在11种芥子油苷,其中脂肪族7种,吲哚族4种。脂肪族和吲哚族芥子油苷分别占总量的63.41%和36.59%。含量最高的为3-丁烯基芥子油苷,最低的为4-甲硫基丁基芥子油苷。The feasibility of glucosinolate quantitative analysis by Sinigrin as external standard and the glucosinolates in bolting stem of Chinese kale were studied. It was found that there was a positive correlation between the concentration of Sinigrin and peak area under the condition of 14.2 U sulfatase and HPLC measurement. The regression equation was y =3E-07x +0.001 and coefficient of regression is 0.9994. The standard recovery and comparison experiments indicated that Sinigrin external standard method is precise and can be used instead of the expensive internal standard Glucotropaeolin. 11 GS compounds in Chinese kale bolting stems were separated by HPLC and confirmed by electrospray mass spectrometry, including 7 kinds of aliphatic GSs and 4 kinds of indolyl GSs which accounted for 63.41% and 36.59% of total GSs content, respectively. Among them, Glucoerucin was detected in Chinese kale for the first time. The highest content GS in bolting stem of Chinese kale was Gluconapin, while the lowest content of GS was Glucoerucin.
