大熊猫核糖体蛋白S12亚基基因(rpS12)的cDNA克隆及序列分析

摘　　要：根据已报道的部分哺乳动物核糖体蛋白S12亚基基因(rpS12)的相关信息设计引物,运用RT-PCR技术,从大熊猫的肌肉组织总RNA中成功克隆了核糖体蛋白S12亚基基因的表达序列,对其进行了克隆、测序及分析.结果表明:大熊猫核糖体蛋白S12亚基基因的表达序列全长为422bp,开放阅读框(ORF)为399bp,编码132个氨基酸的蛋白质,该蛋白的相对分子质量为1.45×104,PI为6.81,含有1个酪蛋白激酶Ⅱ磷酸化位点,3个N-豆蔻酰化位点和一个核糖体蛋白S12 signature位点.该基因的表达序列及其编码的氨基酸序列与已报道的部分哺乳动物有很高的相似性.The cDNA of ribosomal protein S12 gene （rpS12） was cloned successfully from the Giant Panda （Aluropoda melanoleuca） using RT-PCR technology, sequenced, and analyzed. It was found that the cDNA fragment cloned was 422 bp in size, containing one open reading frame of 399 bp. Deduced protein was composed of 132 amino acids with an estimated molecular weight of 1.45 ×10^4and a PI of 6.81. Alignment analysis revealed that both nucleotide and deduced amino acid sequences were highly conserved in five other species also： Homo sapiens, Bos Taurus, Rattus norvegicus, Mus musculus and Sus scrofa. Nucleotide sequence homology of Giant Panda rpS12 to these other species are 95.7%, 96.7%, 92.7%, 93.0%and 6.2% respectively, while amino acid sequence homologies are 100%. Topology prediction showed one casein kinase Ⅱ phosphorylation site, three N-myristoylation sites and one ribosomal protein S12 signature site in the Giant Panda RPS12 protein.
