摘 要:糖蛋白激素受体Lgr5是一种G-蛋白偶联受体超家族的成员,与促甲状腺激素受体、黄体激素受体和卵泡刺激素受体同源。Lgr5多肽链的结构特点为外功能区有17个富含亮氨酸的重复序列,N-端和C-端尾部侧翼富含半胱氨酸序列以及高度保守的7次α螺旋跨膜区。目前尚未发现其生理性配体,故又被称为孤儿受体。Lgr5在人体内分布广泛,大脑、脊髓、乳腺、毛囊、眼睛、生殖器官及胃肠道等都有表达。最近有研究表明Lgr5与干细胞的鉴定和肿瘤的发生密切相关。The glycoprotein hormone receptor Lgr5 is a member of the large G protein-coupled receptors family, which shows high homology with the receptors for TSH£-LH and FSH. The structural character of Lgr5 is that its ectodomains contain 17 leucine-rich repeats together with N- and C-terminal flanking cysteine-rich sequences and it has a highly conserved seven-transmembrane domain. Lgr5 is a kind of orphan receptors due to lack of physiologic ligand. Lgr5 is expressed in diverse tissues including brain, spinal cord, mammary glands, hair folicle, eyes, reproductive organ and so on. Recent studies have shown that Lgr5 is closely associated with the identification of stem cells and tumor formation.
