摘 要:噬菌体展示技术所展示的外源蛋白质可以保持相对独立的空间结构和生物活性,并实现基因和表达蛋白质的体外转换,是蛋白质研究的重要工具。目前用于构建展示文库的噬菌体主要有丝状噬菌体、λ噬菌体、T4噬菌体和T7噬菌体,且在展示功能蛋白结构域、确定核酸结合蛋白质及其相互作用、蛋白质相互作用、蛋白质定向设计和空间结构改造、新受体和配体的发现、筛选酶抑制剂、细胞信号转导、抗原表位分析等方面应用广泛,应用前景良好。Phage display technology is an important tool in protein research, which could maintain the independence between spatial structure and bioactivity of the exogenous protein, and implement the transformation of gene and expressed protein in vitro. Some types of phage, such as λ, T4, T7 and filamentous phage, are used for library construction as main systems. Phage display technology has special advantages and great potentials in displaying of proteins functional domains, determination of nucleic acid binding proteins and the function, protein-protein interaction, directed design and spatial structure reformation of proteins, new receptors and ligands discovery, enzyme inhibitor selection, cell signal transduction research, antigenic epitopes analysis, and so on. All of these decide that the technology has great application prospect in protein research.
