摘  要:目的:以雷公藤多甙建立雌性育龄期小鼠生殖功能低下模型,用于探讨新加归肾丸改善生殖功能低下状态的作用机理。方法:将模型小鼠随机分为5组,每组15只,分别为新加归肾丸高、中、低剂量组、倍美力组、模型空白组,加上正常对照组共6组。连续给药3周,每日定时阴道上皮脱落细胞涂片,观察动情周期变化。第3周末取血、处死小鼠,采用放射免疫法检测血清E2、P、IGF-I水平,组织称重法计算卵巢、子宫指数,病理切片观察卵巢、子宫组织形态学变化。结果:与模型空白组相比,新加归肾丸组动情周期正常小鼠数量增加(P<0.05),卵巢、子宫的组织形态明显改善,高、中剂量组血清E2和P水平明显增高(P<0.01);模型组血清IGF-I与正常组相比显著升高(P<0.01),新加归肾丸各组IGF-I水平与正常组无显著性差异。结论:1、雷公藤多甙可造成雌性小鼠血清IGF-I异常升高,可能为雷公藤多甙致生殖功能损伤的机制之一;2、新加归肾丸能调节IGF-I在卵巢上的适量表达,促进卵泡发育和生长;3、新加归肾丸能改善子宫、卵巢的组织形态,调节卵巢激素水平,使紊乱的动情周期恢复正常。Objective: This study was to establish reproductive hypofunction model of female mature mice by GTW, and investigated the mechanism of improving reproductive hypofunction action of Xinjia Guishen Bill. Methods: The mice were randomLy divided into five groups, 15 in every group, including Xinjia Guishen Bill high, moderate, low - dose group, Premarin (conjugated estrogens tablets) group, model group , and normal control group, the total groups are 6. Successive administrating, smearing vaginal epithelial cell every day on fixture time, observing estrous cycle variation for 3 weeks, and then put all the mice to death. Serum E2, P and IGF- I level were measured by radioimmunoassay. Ovary and uterus index were measured by tissue -weight- assay. Histomorphology of ovary and uterus were observed by pathological slicing. Results: Compared with model group, the number of normal estrous cycle of Xinjia Guishen Bill group were increased ( P 〈 0. 01 ), histomorphology of ovary and uterus were improved too. Serum E2, P level of high, moderate - dose groups were obviously increased ( P 〈 0. 01 ) . Compared with the normal, control group, serum IGF - I level of the model group increased anomaly, but Xinjia Guishen Bill group were normal increased lightly. Conclusion: Xinjia Guishen Bill could regulate some aspects of gonad axis, improve and enhance function of ovary, regulate hormone level in ovary. Thus improve low reproductive function.
