摘 要:目的调查艾滋病病毒Ⅰ型(HIV-1)感染相关的CCR5、CCR2及SDF1等位基因,在广西壮族和汉族普通人群中的多态性及其流行病学意义。方法在广西天等县和南宁市,选取150名壮族和90名汉族健康人作为研究对象,采集外周血,提取基因组DNA,用聚合酶链反应(PCR)扩增CCR5、CCR2及SDF1基因的特定片段。直接根据PCR扩增结果分析CCR5基因多态性,运用限制性片段长度多态性(RFLP)技术分析CCR2及SDF1的多态性。结果全部研究对象的CCR5基因均为野生型,未发现CCR5Δ32突变;CCR2-64I等位基因频率在壮、汉族普通人群中分别为25.7%、26.1%;SDF1-3′A等位基因频率分别为27.7%和27.2%。结论广西壮、汉族普通人群缺乏艾滋病抗性的CCR5Δ32等位基因,而CCR2-64I和SDF1-3’A等位基因频率较高。该研究为深入研究广西壮、汉族普通人群对于HIV-1感染的遗传易感性,以及对艾滋病疫情和病程的影响提供了比较全面、可靠的数据。Objective To investigate genetic polymorphisms and tallele frequencies of CCR5,CCR2,and SDF1 in the general population of Zhuang and Han ethnics living in HIV prevalent areas of Guangxi. Methods Healthy subjects including 150 of Zhuang ethnic and 90 of Han ethnic in Tiandeng county and Nanning city were recruited in the present study. Genomic DNA were extracted from their peripheral blood,and the targeting CCRS, CCR2, and SDF1 genetic fragments were amplified by PCR. CCR5 allele polymorphism was determined by agarose gel electrophoresis of PCR products, while CCR2 and SDF1 allele polymorthisms were analyzed based on PCR-Restriction Fragment- Length Polymorphism(PCR-RFLP). Results All subjects of the two ethnics were homozygous for wild type CCR5, and no subject was found for CCRS-A32 mutation. The CCR2-64I allele frequency in subjects of Zhuang and Han ethnics was 25.7% and 26.1% ,respectively,and the SDF1-3'A allele frequency was 27.7% and 27.2% ,respectively. Conclusion Individuals of Zhang and Han ethnics in Guangxi were in lack of CCRSA32 allele,while the CCR2-64Ⅰ and SDF1-3'A frequencies were relatively high in population of both ethnics. The allele frequencies and the characteristics of gene polymorphisms of CCR5△32, CCR2- 641, and SDF1-3'A would provide basic data for further study of the susceptibility to HIV infection and the impact on prevalence and course of HIV infection among the general population of Zhuang and Han ethnics in Guangxi.
