摘 要:目的:探讨胃康舒宁对胃癌前病变(PLGC)大鼠血清多种癌相关细胞因子的影响.方法:将60只Wistar大鼠随机分为4组:正常组、模型组、维酶素组和胃康舒宁组.采用饮用N-甲基-N-亚硝基-N-硝基胍(MNNG)建立大鼠胃癌前病变模型,用维酶素和胃康舒宁治疗12wk后,用放射免疫法检测各组大鼠血清中表皮生长因子(EGF)、转化生长因子-α(TGF-α)、生长激素(GH)、肿瘤坏死因子-α(TNF-α)、胰岛素样生长因子-Ⅱ(IGF-Ⅱ)、碱性成纤维细胞生长因子(bFGF)的水平.结果:与模型组大鼠相比,胃康舒宁组大鼠血清GH,TNF-α,IGF-Ⅱ水平显著增高(0.24±0.10μg/L vs 0.11±0.04μg/L,91.46±7.63μg/L vs 83.64±3.75μg/L,0.38±0.12μg/L vs 0.27±0.17μg/L,P<0.05或0.01),EGF,TGF-α,bFGF水平明显降低(0.40±0.11μg/L vs 0.56±0.15μg/L,0.021±0.006μg/L vs 0.029±0.012μg/L,1.44±0.37μg/L vs 3.26±0.45μg/L,P<0.05或0.01).结论:胃康舒宁对大鼠胃癌前病变具有良好的治疗和逆转作用,其疗效优于维酶素,而调节EGF,TGF-α,GH,TNF-α,IGF-Ⅱ,bFGF的分泌水平是其防治慢性萎缩性胃炎的作用机制之一.AIM: To investigate the impact of Weikang Shuning Decoction on cytokines involved in the development of gastric precancerous lesions in rats with chronic atrophic gastritis.METHODS: Sixty Wistar rats were randomly divided into four groups, namely, normaI control group, model control group, Vitacoenzyme group and Weikang Shuning decoction group. A rat model of gastric precancerous lesions was created by administration of N-methyl-N-nitroso-N-nitrosoguanidine (MNNG) via drinking water. After the rats were treated with Vitacoenzyme and Weikang Shuning Decoction for 12 weeks, respectively, the levels of serum EGF, TGF-α, GH, TNF-α, IGF-II and bFGF were determined by radioimmunoassay (RIA). RESULTS: The levels of serum GH, TNF-α and IGF-Ⅱin the Weikang Shuning Decoction group were significantly higher that those in the model control group (0.24 ± 0.10 μg/L vs 0.11 ± 0.04 Hg/L, 91.46 ± 7.63 μg/L vs 83.64 ± 3.75 μg/L and 0.38 ± 0.12 μg/L vs 0.27 ± 0.17 μg/L, respectively, P 〈 0.05 or 0.01). In contrast, the levels of serum EGF, TGF-α and bFGF in the Weikang Shuning Decoction group were significantly lower than those in the model control group (0.40 ± 0.11 μg/L vs 0.56 ± 0.15 μg/L, 0.021 ± 0.006 μg/L vs 0.029 ± 0.012 μg/L and 1.44 ± 0.37μg/L vs 3.26 ± 0.45 μg/L, respectively; P 〈 0.05 or 0.01). CONCLUSION: Weikang Shuning Decoction has a good therapeutic effect against gastric precancerous lesions in rats, and its efficacy is superior to that of Vitacoenzyme. Regulation of serum EGF, TGF-α, GH, TNF-α, IGF-Ⅱand bFGF levels may be one of the mechanisms underlying the therapeutic effect of Weikang Shuning Decoction against gastric precancerous lesions.
