摘 要:目的探讨高效抗氧剂α-硫辛酸是否对庆大霉素所致的大鼠耳蜗毒性有防护作用。方法将30只Wistar大鼠随机分成庆大霉素(GM)组、α-硫辛酸(α-LA)+庆大霉素(GM)组及正常对照组,GM组每日腹腔注射GM100mg/kg;GM+α-LA组每日同时腹腔注射GM100mg/kg和α-LA注射液10mg/kg;正常对照组每日腹腔注射生理盐水100mg/kg。3组皆连续用药10d,用药前后测试DPOAE。最后将动物处死,取听泡,观察毛细胞形态。实验数据进行统计学分析。结果DPOAE停药后,GM+α-LA组DPOAE幅值有轻度降低;GM组DPOAE幅值明显降低;2组用药前后比较有显著性差异(P<0.01)。GM+α-LA组大鼠听功能受损程度明显轻于GM组。正常对照组大鼠用药前后DPOAE幅值及形态学无明显变化。形态学观察:给药后GM组耳蜗底回外毛细胞均有较重损伤,前庭毛细胞空泡样改变,纤毛倒伏缺失明显。GM+α-LA组耳蜗底回外毛细胞损伤明显减轻,前庭毛细胞散在缺失。正常对照组耳蜗底回外毛细胞和前庭毛细胞基本正常。结论在使用GM的同时联合应用α-LA可有效减少GM对大鼠的内耳毒性。Objective To investigate protection effect Of a-lipoic acid to entamicin-induced Wistar Rat Cochleotoxicity.Material and methods 30 Wistar rat wererandomly divided into three groups,GM, GM+LA,and Control group.GM group received gentamicin sulfate 100mg / kg daily by intraperitoneal injections; GM+LA group,received gentamicin sulfate 100 mg / kg and injectioa-lipoic acid 10 mg / kg daily by intraperitoneal injections simultaneously; Control group received sterile saline 100 mg / kg by intraperitoneal injections. Duration oftreatment was10 days for all groups.DPOAE measurements were performed twice for each group beforeand the finish of drug administration,Atier the last DPOAE measurement,rat were decapitated immediately, and the temporal bones were removed guickly.The round and oval windows and the apex of the chochlea were opened,perfused with 4 %paraformal-dehyde,and fixed 3h.Aflel'H&E staining. the sections were observed under microscope.The data were analyzed by Student, s Test. Results DPOAE data showed that The amplitudes of DPOAE declined remarkably at any frequency in GM group,DPOAE in GM+LA were also significiantly declined after drug treatment. there was a significiantly difference in GM and GM+LA group (P〈O.05).and the hearing in GM+LA was also better than GM,there was a consistence in morphological changes of them.The morphological results showed that great damage in out hair cell were observed in GM group.and relatively light damage in outer hair cell were observed in GM+LA group.There Was no significant difference in GM+LA and the control group. Conclusion a-lipoic acid,a higher effective antioxidant material can effectiveUy protect GM-induced cochleotoxicity.
