摘 要:对荧光涂料激发能源147Pm辐照人淋巴细胞白血病细胞株Molt-4细胞和巨噬细胞株Ana-1细胞诱发的细胞凋亡进行了研究。实验中估算了147Pm在不同阶段培养细胞中的辐射累积吸收剂量。通过透射电镜的形态观察,发现Molt-4和Ana-1两株免疫细胞在受147Pm辐照时,可诱发明显的核断裂,核染质边聚,以及呈现膜包裹着的凋亡小体形成为特征的细胞凋亡。进而抽提了Molt-4和Ana-1两株免疫细胞的DNA,进行琼脂糖凝胶电泳也显示出阶梯状条带形成的细胞凋亡特征。并且观察到147Pm诱发的免疫细胞凋亡与147Pm辐照的时间和累积吸收剂量相关。The apoptosis in molt-4 cell (an acute human lymphoblastic leukemia cell line)and Ana-1 cell(a macrophage cell line)was studied after irradiation with luminescent paint 147Pm. The cumulativeradiation absorption dose of 147Pm in cultured cells through different period was estimated. Themor-phological changes observed by electron microscope indicated that Molt-4 and And-1 immunecells after exposure to 147Pm demonstrated nuclear fragmentation,margination of condensedchromatin,as well as the membranebound apoptosis bodies formation. The agarose gel electrophoreticobservations showed the DNA ladder pattern formation in Molt-4 cell as well as in Ana-1 cell. Theresults showed that apoptosis induced by 147Pm in immune cells was dependent on the treating timeof 147Pm and cumulative radiation absorption dose.
