摘  要:目的:研究不同药性中药——红参(温性)、生晒参(平性)和西洋参(凉性)对β-淀粉样蛋白(Aβ25-35)诱导SH-SY5Y细胞凋亡的影响。方法:用Aβ25-35处理SH-SY5Y细胞,复制阿尔茨海默病(AD)患者脑内神经元的病理损伤模型,以不同浓度的红参水提物(WERG)、生晒参水提物(WEG)和西洋参水提物(WEAG)进行干预,以倒置显微镜观察其形态学变化;MTT法测定细胞存活率;流式细胞技术检测细胞凋亡率;蛋白印迹法测定兔抗人B细胞淋巴瘤/白血病-2(Bax)、B细胞淋巴瘤/白血病-2连接X蛋白(Bcl-2)的表达。结果:50μmol.L-1Aβ25-35诱导SH-SY5Y细胞72h后,细胞变圆、聚集,细胞存活率显著降低(P<0.01)、凋亡率显著升高(P<0.01)。Aβ25-35与不同浓度的WERG、WEG和WEAG同时孵育后,可显著减少细胞损伤,使细胞存活率升高、凋亡率降低,同时Bax与Bcl-2比值显著降低。结论:不同药性的红参、生晒参和西洋参对Aβ25-35诱导的SH-SY5Y细胞凋亡具有不同程度的保护作用,其中红参作用较强。OBJECTIVE: To investigate the effects of water extracts of Panax ginseng(warm), dry P.ginseng(mild), P.quinque folium (cool) on the apoptosis of SH SY5Y cells induced by β- amyloid protein(Aβ25 -35) .METHODS:SH- SYSY cells were treated with Aβ25- 35 to simulate the pathologic neuronic damage model in patients with Alzheimer's disease, and then treated with different doses of water extracts of P. ginseng(WERG),dry P. ginseng(WEG)or P.quinque folium (WEAG). The morphologic changes of the SH - SY5Y cells were observed by inverted microscope. Cell survival rate was measured by MTT assay. The percentage of apoptosis and the expression of Bax and Bcl - 2 proteins were detected by Western Blot- ting .RESULTS: After treatment with Aβ25 -35 50 μmol · L ^-1 for 72 hours, SH - SY5Y cells became more round and aggregated and the cell survival rate was significantly reduced ( P 〈0.01) but the percentage of apoptosis increased significantly( P 〈 0.01). The concomitant treatment of Aβ25 -35 50 μmol · L ^-1 plus different doses of WERG, WEG or WEAG markedly attenuated cell damage and apoptosis but increase the survival rate of SH - SY5Y cells and markedly reduced the Bax/Bcl - 2 ratio. CONCLUSION: P.ginseng, Dry P.ginseng and P.quinque foliuue have different degree of protective effects on SH- SY5Y cells against apoptosis induced by Aβ25 -35, of the 3 extracts, the P.ginseng is more potent in efficacy than the other two extracts.
