硫酸镁对大鼠继发性脊髓损伤保护作用的实验研究

摘  要:为探讨硫酸镁对大鼠继发性脊髓损伤的保护作用及机制。用30只体重280±20g Wister成年大白鼠随机分为三组:A组(对照组),仅行T10阶段椎板减压;B组(损伤组),C组(治疗组),均行T10阶段椎板减压和Allens重物打击致脊髓损伤,伤后30min时B组腹腔注射蒸馏水1600mg/kg,C组腹腔注射硫酸镁1600mg/kg。48h后切去伤段脊髓组织分别测定H_2O、Ca^(2+)、Mg^(2+)离子含量,观察局部组织病理学改变,超微结构及单位面积的凋亡细胞数。结果显示跟A组比较,B、C组损伤段脊髓组织H_2O、Ca^(2+)含量增多,Mg^(2+)含量减少,组织病理学及超微结构破坏严重,凋亡细胞数升高,而且C组较B组轻。由此可知,病程早期应用硫酸镁可以减轻脊髓损伤后的继发性损伤,从而对受损脊髓起到保护作用。objective to discuss the possible mechanism of protective effects of magnesium sulfate on the injuried spinal cord in rats model. Thirty WisterRats were randomly divided into 3 groups : group A ( Normal group) ; group B (Spinal cord injury) that is SCI;group'C (Treatment group) :SCI with magnesium sulfate treatment. The SCI Animals model was made by using an improved Allen's weight-drop device. After thirty minutes the animals of group B were injected with 1600mg/kg distilled water through Abdominal cavity injection and group C were injected with 1600mg/kg magnesium sulfate. The contents of H2O,Mg^2+ ,Ca^2+ in the injuried spinal cord tissues were examined. The histopathology and uhrastructure of injuried spinal cord tissues were observed through light and electron microscopes. The Apoptosis cell were assayed through TUNEL reagent. The contents of H2O and Ca^2+ in injuried spinal cord tissues in each SCI group were obviously increased compared to those in normal group (P 〈0.61 ) ,and those in group C was less than those in group B( P 〈 0.05). the contents of Mg^2+ obviously decreased (P 〈0.01 ) in the injuried spinal cord tissues. The damage of structure in group C was less serious than in group B. The number of apoptosis cell in group B was more than that in group C (P 〈 0.01 ). Conclussion:Earlly intervention with magnesium sulfate can decreased the secondary spinal cord injury,Thus the injuried spinal cord were protected effectively.
