摘 要:为建立可以检测H9N2亚型流感病毒的蛋白A法免疫传感器,将石英晶片谐振的高灵敏度与免疫反应的特异性相结合,用蛋白A(SPA)在镀金膜的石英晶体电极偶联抗H9亚型流感病毒的单抗构建传感器探头形成免疫传感器。检测结果表明,该方法具有较好的特异性,不与H5亚型禽流感病毒和NDV反应;检测灵敏度达到20个EID50;组成的检测器对相应的流感病毒响应良好,该法与鸡胚病毒分离法检出流感病毒的符合率达到91%。本文结果为检测流感病毒免疫传感器的进一步深入研究奠定了基础。By using the high sensitivity of resonance of quartz crystal and specificity of immunore action, the gold membrane on quartz crystal electrode was coated with SPA, and the monoclonal antibodies against H9 serotype avian influenza virus(AIV) were coupled with the SPA, then the SPA-piezoelectric immunosensor was established and applied for detecting H9 AIV. The detection results showed that the immunosensor displayed better specificity to H9 AIV, and didn't respond to H5 AIV and NDV. The sensitivity test indicated the detecting limit for the H9 AIV could reach to 20 EID50. The accordance rate between the immunosensor and virus isolation by chicken eggs was 91 %. The method provided a foundation for further research on immunosensor for detecting H9 AIV and it could be a new exploration to detect AIV quickly and quantitatively in practical poultry industry.
