摘  要:目的:观测开心散(远志、石菖蒲、人参和茯苓)对血管痴呆性(VD)大鼠学习记忆功能的影响,并探讨该方对VD大鼠海马乙酰胆碱酯酶(AChE)mRNA表达水平的影响。方法:应用永久性结扎双侧颈总动脉造成VD模型;30d后应用Morris水迷宫进行行为学测试;38d后应用RT-PCR半定量的方法检测大鼠海马AChE mRNA的表达水平;应用SPSS13.0软件对实验数据进行统计分析。结果:与模型组相比,开心散组空间探索试验大鼠在目标象限的游泳时间较长,P<0.05;他克林组VD大鼠定向航行试验学习记忆潜伏期较短,大鼠海马AChE mRNA表达水平较低,P<0.05;开心散组大鼠定向航行试验学习记忆潜伏期,空间探索试验目标象限的游泳时间和目标区域的穿越次数,AChE mRNA的表达水平与他克林组无统计学差异,P>0.05。结论:开心散具有改善VD大鼠记忆能力的作用;他克林具有改善VD大鼠学习能力的作用;开心散改善VD大鼠学习记忆的能力与他克林相似,他克林具有抑制VD大鼠海马AChE mRNA表达的作用,开心散改善VD大鼠记忆的机制需要进一步探讨。AIM : To observe the effects of Kaixin Powder ( Radix polygalae, Acorus gramineus soland, Radix et Rhizoma ginseng and Poria) on learning and memory performance and acetylcholine esterase (ACHE) mRNA in Vascular Dementia (VD) rats. METHODS : The rat model of VD was established by ligating both common carotid arteries with silk thread. After 30-day-treatment, their learning and memory were tested by with Morris maze; 38 days later AChE mRNA expression was measured with semiquantitative Reverse Transcription Polymerase Chain Reaction (RT-PCR) method;All the data was analyzed by SPSS 13.0. RESULTS: 1. Compared with model control group, Kaixin Powder group the swimming time was longer in probe task, P 〈 0.05 ; Tacrine group spatial navigation task latencies were shorter, AChE mRNA expression was down-regulated, P 〈 0.05 ; 2. There was no significant difference between Kaixin Powder group and Tacrine group in spatial navigation task latencies, the swimming time and the times through platform zone in the target quadrant in probe task, P 〉 0.05 ; There was no significant difference between Tacrine group and Kaixin AChE mRNA expression, P 〉 0.05. CONCLUSION: Kaixin Powder could enhance VD rats' memory; while Tacrine could enhance VD rats' learning; Kaixin Powder could enhance VD rats' learning and memory as well as Tacrine. Tacrine campus of VD rats, further studies are needed to elucidate VD rats. could down-regulate AChE mRNA expression in hippothe effects of Kaixin Powder on memory performance in
