定期无偿献血者B细胞免疫功能的研究

摘  要:目的:通过对定期无偿全血捐献者、定期无偿血小板捐献者这两个特殊健康人群的B细胞数量以及抗体分泌功能的研究,评价定期捐血对机体体液免疫功能的影响。方法:采用流式细胞术对三组人群(定期无偿全血捐献者、定期无偿血小板捐献者、对照组)外周血中B细胞总数进行数量分析;分离外周血淋巴细胞,加丝裂原美洲商陆(PWM)体外培养10天,取培养上清,ELISA方法检测IgM及IgG的分泌量。采用F检验对三组样本进行方差齐性分析。当P>0.05时认为两个总体的方差相等,用完全随机设计两总体均数比较的t检验进行统计分析。由于Ig分泌量的数值较大,取对数后再做分析。结果:定期无偿全血捐献者及定期无偿血小板捐献者的B细胞数量与对照组的差异无统计学意义(P>0.05);定期无偿全血捐献者及定期无偿血小板捐献者IgM及IgG的分泌量与对照组的差异也无统计学意义(P>0.05)。结论:B细胞的恢复较快,机体在献血间隔期已经将B细胞的免疫功能调节到正常水平。定期献血者能正常发挥其B细胞的免疫功能。Objective: To study the quantity and the function of antibody secretion by B cells in two groups of specialized healthy people who regularly donated whole blood and platelet. To evaluate the effect of regular blood donation as a stimulating factor on humoral immune function. Methods: The number of B cell of three groups of people(regular volunter whole blood donor, regular volunter platelet donor and control group)was quantitatively analyzed by flow cytometry. Lymphocytes were separated from peripheral blood and cultured in 1640 medium with pokeweed mitogen(PWM) for ten days.Then the supernatant was collected and the IgG and IgM were measured by enzyme linked immunosorbent assay(ELISA). The dates from above tests were analyzed by statistics method. Results: The number of B cells between experimental group and control group was showed no difference( P 〉 0.05)and both IgG and IgM secretion by B cells between experimental group and control group were no statistical significance( P 〉 0.05). Conclusion: People adjust the immunological function of B ceils to normal level in blood donation plastochrone. These undoubtedly provide powerfully experimental data on the enlistment of volunter blood donation.
