摘  要:目的探讨聚花过路黄对局灶性脑缺血大鼠脑缺血损伤的保护作用及可能机制。方法采用线栓大脑中动脉法建立雄性Wistar大鼠局灶性脑缺血模型,分成假手术组、模型组、川芎嗪组和聚花过路黄组,手术前2周,灌服蒸馏水或相应药物,观察各组术后缺血3、6、24 h的神经功能评分,检测缺血24 h脑组织含水量、Ca2+含量、脑梗死体积等指标的差异。结果与假手术组比较,模型组大鼠随缺血时间延长,神经功能评分逐渐升高,脑缺血24 h脑组织含水量、Ca2+含量明显升高(P<0.01);缺血24 h后,聚花过路黄组和川芎嗪组大鼠神经功能评分、脑组织含水量、Ca2+含量、脑梗死体积均显著性低于模型组(P<0.01),且聚花过路黄组上述指标均低于川芎嗪组(P<0.05或P<0.01)。结论聚花过路黄对局灶性脑缺血大鼠具有较好的神经保护作用,其作用机制可能与抑制细胞内钙超载有关。Objective To explore the neuroprotective effects and its underlying mechanisms of lysimachia congestifloa hemsl(LCH) on the focal cerebral ischemia in rats. Methods The model of middle cerebral artery occlu- sion in Wistar rats was established by suture method. The animals were divided into sham operated group,model group,ligustrazine group and LCH group. The distilled water or medicine was given respectively by gastric administration for 2 weeks before operation. The neurologic function,water and Ca^2+ contents of cerebral tissue,and infarct size of cerebrum were observed at 3,6,24 hours after cerebral ischemia. Results Compared with sham operated group neural deficient score in rats of model group was increased with the time endurance after ischemia. Meanwhile,water and Ca^2+ content of brain tissue in rats of model group were both elevated(P〈0.01). Compared with model group neural deficient score,water and Ca^2+ contents of cerebral tissue,and infarct size of cerebrum in rats of ligustrazine group and LCH group was decreased significantly at 24 hours after ischemia(P〈0.01). Moreover, the above parameters in rats of LCH group were improved in comparison with those in ligustrazine group (P〈 0. 05,P〈0. 01). Conclusion It is suggested that LCH shows neuroprotective effects of focal cerebral ischemia in rats and its underlying mechanisms might be associated with the inhibition of intracellular calcium overload.
