益气活血复方对兔颈动脉内皮损伤后血管内膜增殖的影响

摘　　要：目的:观察益气活血复方对兔颈动脉内皮损伤后血小板源性生长因子(PDGF)mRNA表达的影响。方法:28只兔被随机分3组:正常组,模型组,益气活血复方组。高脂喂养6周后以球囊拉伤方法建立兔颈总动脉内皮损伤模型。8周时处死动物,以逆转录聚合酶链式反应(RT-PCR)法检测动脉损伤处PDGF-BmRNA表达,HE染色观察颈动脉组织病理形态学改变。结果:与模型组对比,益气活血复方明显抑制了动脉损伤后的内膜增殖反应,下调了内膜损伤处PDGF-BmRNA的表达水平(P<0.05)。结论:益气活血复方可以显著减轻兔血管成形术后内膜增生,其机制可能与抑制PDGF表达有关。Objective： To observe the effects of reinforcing qi activat blood formula （RQABF） on the expression of platelet-derived growth factor （PDGF） in the rabbits after percutaneous transluminal coronary angioplasty （PTCA） with hyperlipidemia. Methods： A total of 28 male New Zealand white rabbits were randomly divided into three groups： The control group, model group and RQABF intervention group. The model of restenosis were established by injuring carotid arteries with PTCA in cholesterol fed rabbits. After 8 weeks, PDGF- 13 measured with reverse transcription polymerise chain reaction （RT-PCR） and pathologic alter were observed with HE staining. Results： By RT-PCR, the expression of PDGF--B mRNA in injured artery were much higher in model group （P〈0.05） , compared with the normal control group, the expression of PDGF-- B and intimal hyperplasia markedly decreased in RQABF group, compared with those of model group （P〈0.05）. Conclusion： Reinforcing qi activat blood formula can inhibit the expression of PDGF--B and lighten the intimal byperplasia after PTCA.
