人参瓜蒌莪术汤对小鼠lewis肺癌细胞增殖的影响

摘  要:目的探讨人参瓜蒌莪术汤对lewis肺癌模型小鼠的治疗作用及机制。方法建立小鼠Lewis肺癌移植瘤模型,分为模型对照组、环磷酰胺对照组和人参瓜蒌莪术汤高、低剂量组。在计算抑瘤率的基础上,运用酶联免疫吸附剂测定法(ELISA)检测瘤组织细胞核抗原(PCNA)和细胞周期蛋白D1(Cyclin D1)的表达。结果人参瓜蒌莪术汤对小鼠Lewis肺癌表现出明显的抑制作用,以中剂量组效果最为明显,抑瘤率可达47.88%;人参瓜蒌莪术汤低、中、高剂量组均能明显降低瘤组织细胞PCNA的表达(P<0.01),中、高剂量组的抑制作用显著强于低剂量组(P<0.01);同时,人参瓜蒌莪术汤低、中、高剂量组均能明显降低瘤组织细胞Cyclin D1的表达(P<0.05),中剂量组效果最为显著,并明显强于环磷酰胺阳性对照组(P<0.01)。结论人参瓜蒌莪术汤具有抑制小鼠Lewis肺癌生长的作用,其作用机制可能与降低肿瘤组织Cyclin D1的表达,抑制瘤细胞的增殖有关。Objective To explore the influence of treatment and mechanism by Renshen Gualou Ezhu Decoction (RGE) to mouse Lewis lung cancer ceils. Methods Based on establishment of international standard model for tumor study - Lewis lung cancer in C57BL/6 Miee model, they ealeulated the inhibition rate and used ELISA to detect the PCNA and Cyclin D1. Fifty C57BL/6 Mice were divided into 5 groups: control group, low- dose RGE group, middle- dose RGE group, high- dose RGE group and Cyelophosphamide(CP) control group. Results Inhibition rate in the middle - dose RGE group reached 47.88%. PCNA expression was reduced on all dose levels - Low, Medium and High. Both medium and high dose groups showed better effect then the low dose group (P 〈 0.01 ). RGE Decoction reduced the Cyclin D1 expression (P 〈 0.05) on all dose levels - Low, Medium, or High. The group with medium dose showed better effect then the CP group (P 〈 0.01 ). Conclusions RGE Decoction can significantly inhibit Mouse Lewis lung cancer cells proliferation. The underlying mechanisms might be related to its reducing the Cyclin D1 expression and resists tumor cell proliferation.
