肾气丸“阴中求阳”配伍对肾阳虚大鼠细胞因子含量的影响

摘　　要：目的用各拆方肾气丸灌胃肾阳虚大鼠,测定灌胃后各组大鼠血清中白细胞介素-1β(IL-1β)、白细胞介素-2(IL-2)、干扰素-γ(IFN-γ)、肿瘤坏死因子-α(TNF-α)的含量,分析肾气丸"阴中求阳"的配伍治疗肾阳虚证的机理。方法以氢化可的松25 mg/(kg.d)注射大鼠,连续12 d,同时以各肾气丸拆方组水煎液灌胃,于灌胃第21天,麻醉大鼠,心脏取血,离心取上清液,用ELISA法检测大鼠血清中IL-1β,IL-2,IFN-γ,TNF-α的含量。结果与正常组比较,模型组IL-1β、IL-2的含量降低,经灌胃后,各拆方组大鼠血浆中二者的含量均有不同程度增加,其中阴阳双补2组含量高于补阳2组,阴阳双补1组含量高于补阳1组,且阴阳双补1组中IL-1β含量高于补阴1组,具有统计学意义。与正常组比较,模型组大鼠血浆中IFN-γ、TNF-α的含量降低,经灌胃后,各拆方组大鼠血浆中二者含量均有不同程度的增加,但增加并不显著。结论肾气丸"阴中求阳"的配伍可能是通过增加血浆中IL-1β、IL-2的含量治疗肾阳虚证,故增加血浆中IL-1β、IL-2的含量,可能是肾气丸"阴中求阳"配伍治疗肾阳虚证的机理之一。Objective To study the effect of the compatibility of Shenqi Pill＂ reinforcing Yang from Yin＂ on the content of cytoline in kidney - Yang deficient rats. Methods The rats were infected with cortisone at the dose of 25 mg/（ kg .d） for 12 days and administered with dismantled decoction at the same time. On the 21th day, the rats were anesthetized and the blood was collect- ed. The contents of IL - 1β, IL - 2, IFN - γ, TNF - α in supernantant were measured by the method of ELISA. Results Compared with normal group, the contents of IL - 1β, IL - 2 were lowered in model group. After intragastric administration, the contents of IL - 1β, IL - 2 were increased at different levels in rat plasma of each dismantled group, and the contents of IL - 1β, IL - 2 in the second group of invigorating Yin - Yang was higher than those in the second group of invigorating Yang, The first group of invigorating Yin - Yang was higher than that in the first group of invigorating Yang. The contents of IL - 1β in first group of invigorating Yin - Yang is higher than the first group of invigorating Yin. And all had statistical significance. Compared with normal group, the contents of IFN - γ, TNF - α were reduced in model group. After intragastric administration, the contents of IFN - γ, TNF - α were increased at different levels in plasma of each group rats. Conclusion The increase of content of IL - 1β, IL - 2 in plasma is likely to be the mechanism that Shenqi Pill treat the kidney - yang asthenia syndrome by the compatibility of ＂ reinforcing Yang from Yin＂.
