健脾活血方对2型糖尿病大鼠肝细胞膜胰岛素受体的影响

摘　　要：目的探讨健脾活血方对2型糖尿病胰岛素抵抗(Insulin Resistance,IR)大鼠肝细胞膜胰岛素受体(Insulin Recep-tor,InsR)mRNA表达的改善作用及机理。方法取Wistar大鼠60只,雌雄各半,采用链脲佐菌素(Streptozotocin,STZ)加高脂饮食造成2型糖尿病胰岛素抵抗大鼠模型。设空白组、模型组、健脾活血方低剂量组、健脾活血方高剂量组和罗格列酮组,以RT-PCR法检测大鼠肝细胞膜InsR mRNA的表达。结果健脾活血方高剂量组大鼠肝脏InsR mRNA的表达与罗格列酮组相当,与模型组比有显著性差异(P<0.01)。结论健脾活血方能够上调大鼠肝细胞膜InsR mRNA的表达,从而改善胰岛素抵抗。Objective To explore the effect of Strengthening Spleen and Activating Blood Prescription on expressions of insulin receptor gene in the liver membrane of Type 2 Diabetes Mellitus rats and its mechanism. Methods 60 Wistar rats , half male and half female, were randomly devided into five groups： normal control group, model group, Strengthening Spleen and Activating Blood Prescription lowdose group and highdose group, and Rosiglitazone group. The model of insulin resistance of type 2 diabetebes mellitus was established by STZ injection and high - fat diet. Expression of insulin receptor gene in liver membrane was detected. Results The expressions of liver insulin receptor gene in Strengthening Spleen and Activating Blood Prescription high dose group was the same with Rosiglitazone group. There were significant differences compared with model group （P 〈 0.01 ）. Conclusion Strengthening Spleen and Activating Blood Prescription can up - regulate the expressions of liver insulin receptor gene in type 2 diabetic rats, and improve insulin resistance.
