摘 要:将水培65 d的烟株进行打顶处理,测定打顶和不打顶烟株上部叶的烟碱含量。分别提取根中总RNA,利用腐胺-N-甲基转移酶(PMT)基因特异引物进行actin做内参的半定量RT-PCR,分析打顶前后PMT转录水平。结果表明,上部烟叶烟碱含量和腐胺-N-甲基转移酶基因表达量的变化趋势是一致的。The tobacco plants were topped after being water - cultivated for 65 days, and the nicotine content in the top leaf of topped and untopped tobacco plants was examined. Total RNA was extracted respectively from the root of topped and untopped tobacco plants. Expression analysis of putrescine N -methyl transferase (PMT) was carried out by semi -quantitative RT -PCR with the tobacco actin gene as native control. Results showed that the changeable tendency of nicotine content in the top leaf and PMT expression was consistent.
