摘  要:目的研究一种改良的从全血中分离中性粒细胞的方法。方法本研究改进了细胞分离装置,采用人血浆稀释高密度Percoll液,配制成1.077g/ml和1.090g/ml两种等渗Percoll不连续密度梯度分离液,分离人血,得到纯中性粒细胞。结果经瑞士-吉姆萨染色和台盼蓝染色证明细胞纯度达97.7%,存活率在97%以上。通过细胞的瑞士-吉姆萨染液染色涂片对比,比较了不同方法分离的细胞的形态,本方法分离的细胞外观形态与细胞原始形态最接近。结论与国内外的分离方法相比较,该方法简便易行,且分离纯度和细胞活性高,尤其是对细胞形态影响小,是一种高效、经济的分离方法。Objective To study an improved method of separating neutrophils from whole blood. Methods The cell separation device was improved, and plasma was employed to dilute the high-density Percoll liquid to prepare two kinds of separation liquids with 1.077g/ml and 1.090g/ml for iso-osmotic Percoll discontinuous density gradient, which was applied to separating neutrophils from human blood. Results The cellular purity tested by Wright-Giemsa staining was 97%, and the survival rate tested by Trypan blue staining was more than 97%. The morphology of cells separated by different methods was compared by doing smears contrast after Wright- Giemsa staining, the ceils separated with this method was most like their original morphology. Conclusions Compared with other domestic and foreign separation methods, this method is easy to implement, and both cell separation purity and cell activity show a high level, furthermore, it has little influence on cellular morphology. It is a highly efficient and economical method for cellular separation.
