摘 要:目的:观察垂盆草醇提物(CCW)对人肝癌细胞HepG2生长及其肿瘤相关分子指标的影响。方法:MTT法测定CCW对HepG2细胞增殖的作用;流式细胞术观察细胞周期的改变;分别用RT-PCR及免疫细胞化学法检测CCW对VEGF、c-Myc基因及蛋白水平的影响。结果:HepG2细胞的增殖在CCW作用的第24、48和72 h均被显著地抑制;CCW(100、200μg.m l-1)可使HepG2细胞阻滞于G0/G1期;CCW可显著下调VEGF、c-Myc的mRNA及蛋白水平的表达。结论:CCW对HepG2细胞增殖具有明显的抑制作用,并且能阻止细胞进入G2/M期,这可能与c-Myc基因表达的下调密切相关;CCW能抑制HepG2细胞的VEGF分泌,初步推测CCW具有抗血管生成作用。Objective To investigate the effect of ethanol extracts from Sedum sarmentosum Bunge (CCW) on HepG2 cells' growth and the ehanges of their tumor-associated molecular targets. Methods Firstly proliferation abil- ity and cell cycle changes of HepG2 cells were detected by MTY method and flow cytometry. The mRNA levels of VEGF and c-Myc were determined by RT-PCR. The protein expressions of VEGF and c-Myc were valued by immunocytochemis- try. Results CCW greatly inhibited HepG2 cell growth after CCW treatment for 24,48 and 72h. Analysis of cell cycles demonstrated CCW caused HepG2 cells arrested in G0/G1 phase. The mRNA and protein levels of VEGF and c-Myc were significantly decreased after CCW treatment for 48 h. Conclusion The present study suggests that CCW have potential inhibition effects on HepG2, which is in close correlation with the declines of c-Myc transcripts and protein translation in HepG2 cells. It also presumed that CCW might possess anti-angiogenic activity by inhibiting secretion of VEGF.
