摘  要:运用电镜酶细胞化学技术显示,罗氏沼虾经高浓度Cu2+(5.12 mg/L)处理一周后,体内3种组织(鳃、肝胰腺和心脏)细胞超微结构均有不同程度的损伤,主要表现为细胞核收缩,线粒体膨胀和内嵴减少,细胞结构模糊、胞质浑浊,髓样小体增多,脂滴破裂等;超氧化物歧化酶(SOD)阳性颗粒在健康虾中主要存在于细胞核、线粒体膜周围和脂滴周围等,Cu2+胁迫后,组织细胞中线粒体膜和脂滴周围的SOD阳性颗粒明显减少,细胞核内SOD阳性颗粒缺失.结果表明:高浓度Cu2+对罗氏沼虾体内3种组织细胞中SOD的表达具有显著抑制作用,可导致虾体免疫机能显著下降,从而出现组织细胞结构明显损伤.Enzyme cytochemical technique of electronic microscope was used to observe adult Macrobrachium rosenbergii which were raised in the breeding water containing 5.12 mg/L Cu^2+ for one week. The result showed that, being treated by high concentration of Cu^2+, the three tissues ( gill, liver and heart) chosen from Macrobrachium rosenbergii were damaged in different ways, which could be concluded as follows: the deflation of nucleus, the inflation of mitochondria and less mitochondrial cristaes, unclear cell ultrastructure and cytoplasm, more myelin figures, cracked lipid droplets. SOD positive granules were mainly found around nucleus, mitochondrial membrance and lipid droplets in healthy Macrobrachium rosenbergii. While in Cu^2+ treated subjects, the SOD positive granules around mitochondrial membrance and lipid droplets reduced and they missed in nucleus. All that above suggested high concentration of Cu^2+ (5.12 mg/L) gave a evident inhibition on the expression of SOD in all the three tissues which may cause decrease in immunity function and obvious damage in cell ultrastructure.
