摘 要:目的从海洋链霉菌GB-2的发酵液中分离纯化抗真菌物质,并研究该物质的稳定性。方法采用有机溶剂萃取,硅胶层析和制备型高效液相色谱(HPLC)等分离技术纯化抗真菌物质;通过管碟法,以米曲霉为指示菌研究抗真菌物质的pH、温度和紫外稳定性。结果海洋链霉菌GB-2产的抗真菌物质得到了纯化;该物质在pH1.0、pH13.0、100℃和紫外照射24h条件下抑菌活性几乎不变。结论纯化了海洋链霉菌GB-2抗真菌物质具有很好的稳定性,为其结构鉴定和开发应用提供了理论依据。Objective The purification and stability of antifungal substances produced by Streptomyces sp. GB-2 were studied. Methods The antifungal substances were separated and purified by organic extraction, silica gel chromatography and preparative high performance liquid chromatography(HPLC) ; take Aspergillus oryzae as indicating fungus,the stabilities of the antifungal substances under different conditions of pH, temperature and UV were studied by cylinder-plate method. Results The antifugual substances were purified and the stability analyses of the extracts from Streptomyces GB-2 fermentation broth showed very stable at pH 1.0, pH13.0, 100℃ and changed very little under UV treatment for 24 hours. Conclusion The purified antifungal substances showed good stability which provided theoretical foundation for their structural identifications and future applications.
