摘 要:采用控制加权可变步长单纯形算法对PTC氨基酸的毛细管胶束电动色谱分离体系进行动态优化。寻优过程中同时考虑背景电解质的pH值、十二烷基硫酸钠浓度和有机添加剂浓度对体系分离效果的影响。分别使用Long系数法和均匀设计表法确定初始单纯形顶点对分离体系进行动态优化。优化结果使17种PTC氨基酸获得了比较满意的分离。The systematic optimization for the MEKC separation of PTC amino acids by using a dynamic scouting optimization method controlled weighted centroid variable size simplex algorithm was established. The optimization procedure simultaneously concerned the pH value of the background electrolyte (BGE), the sodium dodecyl sulphate (SDS) concentration and the percentage of organic solvent. These influence factors were complicated and nonlinear with each other, and the normal optimization strategy using single factor method was not able to achieve the optimum separation conditions practically. The controlled weighted centroid variable size simplex algorithm possessing the characteristics of simple experimental design and dynamic scouting optimization was not limited by the number and the complex mutual relation of the influence factors. Two methods, the Long Coefficient and Uniform Design Table, were used to define the start vertexes during the optimization procedure. The similar results of dynamic scouting optimization were obtained with use of either of the two methods to define the start vertexes. The satisfactory separations( m p=17, R S m ≥3 82)were gained at pH from 7 05 to 7 40, SDS concentration from 36 70 to 40 10mmol/L and isopropanol concentration from 127 0 to 140 0g/L.
