摘  要:目的观察重组人促红细胞生成素(r-HuEPO)对颅脑损伤后大鼠脑组织中抗凋亡因子X连锁凋亡抑制蛋白(XIAP)和促凋亡因子第二个线粒体衍生的半胱氨酸蛋白酶激活剂(Smac/DIABLO)表达的影响,并探讨二者在r-HuEPO抗凋亡中的作用。方法采用自由落体法建立大鼠颅脑损伤模型,在伤后不同时间点用末端标记原位细胞凋亡检测法(TUNEL法)检测细胞凋亡,用免疫组织化学法检测XIAP和Smac/DIABLO表达的变化。结果与假手术组相比,颅脑损伤后打击周边区的细胞凋亡数显著增加(P<0.01),XIAP和Smac/DIABLO表达水平明显升高(P<0.05);与对照组相比,r-HuEPO治疗组XIAP的表达在2~72 h时间点明显升高(P<0.05),Smac/DIABLO的表达和细胞凋亡数在6h至7d时间点显著下降(P<0.05)。结论r-HuEPO可能通过促进XIAP的表达和抑制Smac/DIABLO的表达减少颅脑损伤后的细胞凋亡,发挥脑保护作用。Objectives To explore the effect of recombinant human erythropoietin (r-HuEpo) on the expressions of X chromosome linked inhibitor of apoptosis protein (XIAP) and the second mitochondria-derived activator of caspase (Smac/DIABLO) in the brain tissues in rats with traumatic brain injury (TBI) and the roles of XIAP and Smac/DIABLO in the resistance of r-HuEPO to apoptosis. Methods The experimental TBI model was established by Feeney's method. The apoptosis of cells and expressions of XIAP and Smac/DIABLO in the injured cerebral cortex tissues which were obtained at different time after TBI were determined respectively by TUNEL and immunochemical method. Results The number of apoptotic cells and the expressions of XIAP and Smac/DIABLO increased significantly in TBI group compared to those in sham group (P〈0.05). The expression level of XIAP was significantly higher 2, 6, 24 and 72 hours after TBI in r-HuEPO group, and the expression level of Smac/DIABLO and the number of apoptotic cells were significantly lower 6, 24, 72 and 168 hours, after TBI in r-HuEPO group than those in TBI group (P〈0.05). Conclusions R-HuEPO may inhibit neuronal apoptosis after TBI through enhancing expression of XIAP and suppressing expression of Smac/DIABLO; it may plays a neuroprotective role in the brain tissue after TBI thereby.
