摘  要:目的观察三氧化二砷(As2O3)对293T细胞凋亡和增殖的影响并探讨SHP-2在其中的作用。方法显性负性SHP-2突变体SHP-2C459/S与具有绿色荧光蛋白基因的载体pEGFP-C1共转染入HEK293T细胞,Ho-echst33258染色检测细胞凋亡,细胞计数及荧光分光光度法检测细胞增殖。结果SHP-2C459/S突变体能增强细胞凋亡和减轻增殖抑制。结论SHP-2可能参与到As2O3诱导的细胞凋亡和增殖抑制的信号转导通路中。Objective To investigate potential role ( As2O3 ). Methods Dominant negative SHP-2^C459/S of SHP-2 in apoptosis induced by arsenic trioxide mutant was cotransfected with a vector harbouring green fluorescent protein gene pEGFP-C1 (as the marker gene) into HEK293T cells. Hoechst33258 staining were used for apoptosis assays, and cell counting and fluorescence spectrometry were used for proliferation assays. Results The introduction of SHP-2^C459/S mutaht increased the apoptosis and decreased the proliferation inhibition. Conclusion SHP-2 may involve in the pathways of apoptosis and prolifera- tion inhibition induced by As2O3.
