摘  要:目的探讨葡萄籽提取物原花青素(grape seed procyanidin extract,GSPE)对人膀胱癌BIU87细胞周期的影响及其可能机制。方法应用不同浓度GSPE(50、100、200μg/ml)与膀胱癌BIU87细胞共同培养24 h后,采用流式细胞术检测细胞周期,RT-PCR、Western blot检测CyclinD1、CDK4mRNA及蛋白表达变化。结果GSPE可呈剂量依赖性阻滞细胞于G1期,并可引起CyclinD1和CDK4mRNA及蛋白水平的表达减少(P<0.01)。结论GSPE可诱导人膀胱癌BIU87细胞周期阻滞,其机制可能是通过下调CyclinD1和CDK4的表达而实现。Objective To study the effect of grape seed procyanidin extract (GSPE) on the cell cycle arrest in human bladder cancer BIU87 cells and investigate the underlying molecular mechanism. Methods BIU87 ceils were treated with different concentrations of GSPE (50, 100 and 200 μg/ml) and cultured for 24 h. The untreated group served as control. Flow eytometry was used to evaluate the cell cycle, and RT-PCR and Western blotting were used to detect the mRNA and protein expressions of CyclinD1 and cyclin-dependent kinase 4 (CDK4 ). Results GSPE inhibited the cell growth through cell cycle arrest at G1 phrase in a dosedependent manner. Semi-quantitated RT-PCR and Western blot analyses indicated that GSPE decreased CyclinD1 and CDK4expressions in a dose-dependent fashion (P 〈 0.01 ). Conclusion GSPE induces cell cycle arrest in BIU87 cells, and the effect may be related with its down-regulation of CyclinD1 and CDK4.
