摘  要:目的通过观察人参二醇组皂苷(PDS)对TRPV6蛋白的影响,探讨PDS对LPS诱导休克时脑损伤的保护机制。方法将大鼠随机分为对照组、LPS休克(LPS)组、地塞米松(LPS+Dex)组、PDS低剂量(LPS+PDSL)组、PDS中剂量(LPS+PDSM)组及PDS大剂量(LPS+PDSH)组。大鼠静脉注射LPS(4 mg/kg)4 h后,观察各组大鼠生存期,计算生存率,Western Blot检测大脑皮质中TRPV6和Bcl-2的蛋白表达。结果与对照组相比,LPS组大鼠生存率降低达50%,脑皮质中TRPV6的蛋白表达水平明显降低,Bcl-2/Bax蛋白表达的比值降低;而与LPS组相比,LPS+Dex组和LPS+PDS各组大鼠生存率增加,脑皮质中TRPV6的蛋白表达水平明显增高,Bcl-2/Bax比值也增高。结论PDS减轻LPS诱导的脑损伤,可能与上调脑皮质中TRPV6蛋白有关。Objective To explore the protective effect of panaxadiols(PDS) on brain injury induced by lipopolysaccharides(LPS) and its mechanism.Methods Rats were divided into control,LPS,LPS + dexamethasone(DEX) and LPS + PDS low, middle and high dose groups randomly.Survival rate of rats in every group was calculated.The expressions of TRPV and BCL-2 were detected by Western blot. Results Compared with control group,survival rate of LPS group was decreased,the expression of TRPV6 and BCL-2/BAX protein ratio were decreased.Compared with LPS group,survival rates of LPS+DEX group and LPS+PDS groups were increased,and the expressions of TRPV6 protein and BCL-2/BAX protein ratios were also upregulated in LPS+Dex group and all LPS+PDS groups.Conclusions The effect of PDS against brain injury induced by LPS may be related with upregulating TRPV6 protein.
