禽腺病毒Ⅰ型琼脂扩散抗原的研制及应用

摘　　要：研制了禽腺病毒Ⅰ型琼脂扩散抗原,检测了该抗原的效价和特异性,并用该抗原对人工感染鸡沉淀抗体的变化以及现地血清样本进行了检测。结果显示,该抗原不与其他主要禽类病原体阳性血清反应,可与1∶8稀释的禽腺病毒Ⅰ型阳性血清反应。对3月龄SPF鸡进行人工感染后,第7 d可检测到沉淀抗体,血清中的沉淀抗体可持续3个月以上。抗原在-20℃条件下可保存2年。经对现地38个鸡场进行检测,其中阳性场14个,阳性率为36.84%,表明我国的禽腺病毒感染率很高。结果证实,研制的禽腺病毒Ⅰ型琼脂扩散抗原敏感、特异,可用于禽腺病毒感染的血清学检测。Fowl adenovirus typeⅠ（FAV-Ⅰ） agar diffusion antigen was developed,and the titer and specificity of the antigen was detected.Precipitating antibody changes of chickens after virus inoculations and some field serum samples were detected with the antigen.The antigen did not react with the other major chicken pathogen positive sera.It reacted with FAV-Ⅰ-positive serum diluted at 1∶8.The precipitating antibody was detected in the sera of SPF chickens inoculated with FAV-Ⅰ CELOV strain on day 7 post-inoculation,and it lasted for more than three months.The antigen could be stored for two years at-20 ℃.Infection rate of fowl adenovirus was 36.84%（14 out of 38 field farms） indicating that infection rate of avian adenovirus in China was very high.The results showed that the developed FAV-Ⅰ agar diffusion antigen was sensitive and specific,and could be used for serological diagnosis of fowl adenovirus infection.
