含二苯乙烯苷大鼠血清对NG108-15痴呆模型细胞的影响

摘　　要：目的:观察含二苯乙烯苷(TSG)血清对NG108-15痴呆模型细胞的影响。方法:分别用TSG(0.3g·kg-1)、阳性对照药石杉碱甲(0.02mg·kg-1)等灌胃大鼠10d后分离相应的含药血清,以β淀粉样蛋白(Aβ)25-35诱导NG108-15细胞成为痴呆细胞后用相应的含药血清培养,观察NG108-15细胞的生长状态、存活率、突起率及突起平均长度,并与模型组比较。结果:无论在增殖相或分化相中,TSG组细胞生长状态均良好;在增殖相细胞中,TSG组、模型组存活率分别为89.1%、64.6%(P<0.01);在分化相细胞中,TSG组存活率为74.3%,细胞突起率为(50.83±6.66)%,突起平均长度为(21.52±5.19)μm,模型组分别为59.2%、(41.42±7.23)%、(15.59±4.54)μm(P<0.05或P<0.01),TSG组各指标与石杉碱甲组相近。结论:含TSG大鼠血清对Aβ25-35诱导的NG108-15痴呆模型细胞损伤具有保护作用。OBJECTIVE： To evaluate the effect of stilbene glucoside （TSG）-containing serum on NG108-15 cells model of Alzheimer＇ s disease （AD） . METHODS： The rats were intragastrically administered with either TSG 0.3 g·kg^-1 or huperzine A （positive drug, 0.02mg·kg^-1） for 10 days, then the drug-containing serum were sampled from the rats. NG108 - 15 cells were induced into model cells of AD by Aβ 25 - 35, and the model cells of AD were cultured with corresponding drug - containing serum. The morphology, survival rate, neurite outgrowth and average length of outgrowth of NG108- 15 cells were observed and compared with the model group. RESULTS： Whether in the proliferation phase or differentiation phase, the cell growth was in good condition in TSG-treated group ; in the proliferation phase, the survival rate was 89.1% in TSG-treated group versus 64.6% in model group （P〈0.01）. In the differentiation phase, the survival rate, the neurite outgrowth and the average length of outgrowth were 74.3%, （50.83±6.66）% and （21.52±5.19）μm, respectively in TSG-containing serum group versus 59.2%, （41.42±7.23） % and （15.59±4.54）μm, respectively in model group （P 〈 0.05 or P〈0.01） . The indexes in TSG group were similar to those in the huperzine A group. CONCLUSION： TSG- containing rat＇ s serum has protective effect on the injury of NG108- 15 ceils of AD induced by Aβ 25- 35.
