摘  要:目的:为研究中药复方防治艾滋病的机制,探讨湘A1号、2号对免疫缺陷模型小鼠的T细胞亚群的影响。方法:经腹腔注射环孢菌素A建立以CD4+T细胞数目减少为主要特征的免疫缺陷小鼠模型。试验设正常对照组,模型对照组,阳性药物组,湘A1号低、中、高剂量观察组,湘A2号低、中、高剂量观察组,每组10只。灌胃给药,每次0.7 ml/只,2次/d,连续给药1周,最后一次给药后禁水、禁食8 h,取标本进行T细胞亚群检测。结果:与模型对照组比较,湘A1号中、高剂量能显著上调CD4+T细胞亚群的百分率(P<0.01或P<0.05),低、中、高剂量组能下调CD8+T细胞亚群百分率(P<0.01或P<0.05),调整CD4+/CD8+T细胞比值(P<0.01或0.05);湘A2号中、高剂量组能显著上调CD4+T细胞亚群的百分率(P<0.01),下调CD8+T细胞亚群的百分率(P<0.01或P<0.05),调整CD4+/CD8+T细胞比值(P<0.01或0.05),其中中剂量组对CD8+T细胞亚群百分率的下调作用和对CD4+/CD8+T细胞比值调整作用的效果优于对照药物。结论:湘A1号、2号能调节环孢菌素A所致的免疫缺陷动物模型CD4+、CD8+T细胞亚群的百分率和CD4+/CD8+T细胞比值。Objective: To study the mechanism of the Chinese medicinal compound in the prevention and treatment on AIDS, and discuss the effects of the two Chinese medicinal compounds Xiang A1 and Xiang A2 on the T-lymphocyte subsets in the mouse model of immunodeficiency. Methods : By intra- peritoneal injection of cyclosporine A, we established the immunodefieieney model in 80 mice with the main feature of reduced number of CD4^+ T cells, and equally divided them into 8 groups : model control, positive medication, low-dose Xiang A 1 (L-XA1), medium-dose Xiang A 1 (M-XA1),high-dose Xiang A1 ( H-XA1 ) , low-dose Xiang A 2 (L-XA2) , medium-dose Xiang A and high-dose group Xiang A 2 ( H-XAH), in addition to a normal control group. Medic ducted intragastrically at the dose of 0.7 ml per mouse and twice a day for 7 days. After the mice were fed on nothing for 8 hours, and then their spleens were harvested for the lymphocyte subsets. Results: Compared with the model control group, the M-XA1 and 2 ( M-XA2), ation was conthe last dose, detection of T H-XA1 groups showed a significant increase in the percentage of CD4^+ T cells (P 〈 0. 01 or 0. 05 ), while the L-XA1, M-XA1 and H-XA1 groups exhibited an obvious decrease in that of CD8 ^+ T cells (P 〈 0.01 or 0.05) , with the CD4^+/CD8 ^+ ratio significantly upregulated (P 〈 0.01 or 0.05). Mediumand high-dose Xiang A2 significantly lifted the percentage of CD4^+ T cells ( P 〈 0.01 ), reduced that of CD8 ^+ Tcells (P 〈 0.01 or0.05), and upregulated the CD4^+/CD8 ^+ ratio (P 〈 0.01 or 0.05). Medium-dose Xiang A1 and 2 produced better effects in decreasing the percentage of CD8 ^+ T cells and increasing the ratio of CD4^+/CD8 ^+ in the spleen tissues as compared with the mode] control group. Conclusion : Xiang A 1 and 2 can regulate the percentage of CD4^+ and CD8 ^+ T-lymphocyte subsets as well as their ratio in mice with cyclosporine A induced immunodeficiency.
