摘 要:目的:低氧预适应可使重要脏器对缺血缺氧产生耐受,是一种内源性保护现象。文中探讨低氧诱导因子-lα(HIF-lα)和血管内皮生长因子(VEGF)在移植肝肝内胆管上皮细胞的表达情况及引起移植肝胆管并发症的因素。方法:建立大鼠自体肝移植模型,AT组为自体肝移植,HP组为低氧预适应后行自体肝移植,并设正常对照组。于术后6、12、24、48 h取材,光镜下观察胆管上皮细胞的病理变化,免疫组织化学法检测各组胆管上皮细胞HIF-lα、VEGF的表达情况。结果:光镜下与HP组相比,AT组胆管上皮细胞损伤严重,细胞排列缺失明显,炎性细胞浸润增加;各组胆管上皮细胞均有HIF-lα、VEGF的表达,其中HP组表达最强。结论:HIF-lα、VEGF可能参与了低氧预适应对移植肝肝内胆管上皮细胞的保护作用。Objective: Hypoxic preconditioning (HP) is referred as an endogenous protective process of important organs, which can obtain tolerance of hypoxia and ischemia. This paper is to investigate the expressions of the hypoxia-indueible faetor-1α (HIF-1α) and the vascular endothelial growth factor (VEGF) in intra-hepatic cholaniocytes, and the factor of biliary complications after liver graft. Methods : We established the model of autologous liver transplantation, included 3 groups in this study : an AT ( autologous liver transplantation), an HP (bypoxic preconditioning) and an NC (normal control) group. We observed the pathological changes of the cholaniocytes under the light microscope at 6, 12, 24 and 48 hours after operation, and detected the expressions of HIF-1α and VEGF by the immunohistochemical method. Results : The cholaniocytes in the AT group showed serous damage, increased inflammatory infiltration and obvious absence of orderly arrangement as compared with the HP group. The expressions of HIF-1α and VEGF were higher in the whole process in the HP than in the AT group. Conclusion: HP has a protective effect on cholangiocytes after liver graft, and VEGF and HIF-1α may play an important role in this process.
