用原位杂交检测基因枪轰击小鹅瘟病毒VP3基因疫苗在小鼠体内分布规律

摘　　要：将小鹅瘟病毒(Goose Parvovirns,GPV)VP3基因疫苗(pcDNA-GPV-VP3)分别按每只6、3、1μg的剂量基因枪轰击免疫4周龄BALB/c小鼠,于免疫后不同时间采集小鼠心、肝、脾、肺、肾、肠、脑和免疫部位皮肤等组织,用原位杂交方法检测pcDNA-GPV-VP3在小鼠体内的分布规律。结果显示,pcDNA-GPV-VP3在免疫后1h即分布于小鼠除脑外各个组织,脑组织于免疫后3h呈阳性,各组织阳性信号随时间的推移逐渐减弱;pcDNA-GPV-VP3在不同组织中持续时间依次为肝脏>心>脾>免疫部位>肾、肠>肺>脑;pcDNA-GPV-VP3不同剂量组信号强弱和持续时间存在的总体规律依次为6μg组>3μg组>1μg组。基因枪轰击pcDNA-GPV-VP3于免疫后能快速而广泛的分布于小鼠各个组织且持续存在达63d。Four-week-old BALB/c mice were injected via gene gun bombardment with different doses （6, 3 and 1μg） of the pcDNA-GPV-VP3 vaccine. In situ hybridization was used to detect the distribution of the pcDNA-GPV-VP3 in the organs （hearts, livers, spleens, lungs, kidneys, brains, intestines and skins of inoculation sites） at different time points post injection. The pcDNA-GPV-VP3 could be detected in all tissues at lh post administration but brains became positive at 3h. The positive signals in all tissues decreased afterwards. The duration time in all tissues was in order： liver〉heart〉spleen〉skin〉kidney and intestine〉lung〉brain. The signal intensity and duration time of different doses were： 6μg group〉3μg group〉1μg group. These results demonstrated that pcDNA-GPV-VP3 could quickly distribute in all tissues of mice and exist for at least 63 days post administration.
