摘 要:采用体外培养方法和放射免疫分析法(RIA)研究磷酸二酯酶5(PDE5)抑制剂Sildenafil(西地那非)对小鼠卵母细胞体外自发成熟的作用。结果显示:该抑制剂显著抑制(P<0.05)小鼠卵母细胞的自发成熟,且其抑制作用是可逆的。放射免疫分析实验表明,Sildenafil对卵丘细胞(CCs)和裸卵母细胞(DOs)内环鸟苷一磷酸(cGMP)水平没有显著的升高作用,却能够显著提高CCs和DOs内环腺苷一磷酸(cAMP)的水平。以上结果提示,cGMP和cAMP信号通路之间的互作可能参与了PDE5调控的小鼠卵母细胞自发成熟过程。The objective of the present study was to investigate the effects of PDE5 inhibitor Siidenafil on mouse oocyte spontaneous maturation and to explore the mechanisms by which PDE5 influenced eocyte spontaneous maturation. Oecytes maturation was evaluated through in vitro culture and intracellular cGMP and cAMP levels in cumulus cells (CCs) and denuded oocytes (DOs) were determined by Radioimmunoassay (RIA). The results Showed that Sildenafil (1 mol/L) significantly inhibited spontaneous maturation of cumulus-oocyte complexes (COCs) and that the inhibitory effects of Sildenafil were reversible. The data of RIA revealed that cGMP levels in DOs and CCs did not significantly increase in response to 1 mol/L Sildenafil. In contrast, Sildenafil ( 1 mol/L) was found to remarkably promote cAMP levels both in CCs and DOs at the end of culture. These results suggested that an interaction between cGMP and cAMP signal might be involved in PDE5-mediated mouse cocyte spontaneous maturation.
