摘 要:目的分析高脂血症大鼠二磷酸腺苷(ADP)和花生四烯酸(AA)诱导的最大血小板聚集率(MPAR)、P选择素(P-Selectin)、血栓素B2(TXB2)、6酮前列腺素F1a(6-k-PGF1a)、TXB2/6-k-PGF1a(TP)比值、血小板表面CD62P表达的变化,及上述各指标的相关性。方法24只Wistar大鼠随机分为常规饲料组、高脂饲料组,分组进行干预8周后检测两组大鼠ADP、AA诱导的MPAR,应用酶联免疫法测定大鼠血浆中P-Selec-tin、TXB2、6-k-PGF1a质量浓度,应用流式细胞技术(flowcytometric,FCM)检测血小板表面CD62P的表达,并进行统计学分析。结果高脂干预后,高脂饲料组大鼠ADP及AA诱导的MPAR、P-Selectin、TXB2、6-k-PGF1a、TP比值、血小板表面CD62P的表达分别为(55±14)%、(30±8)%、(27.0±2.8)μg/L、(54±9)μg/L、(12.6±2.8)μg/L、(3.25±0.61)%、(25.2±5.3)%,与常规饲料组的(24±5)%、(17±3)%、(10.7±1.7)μg/L、(14±4)μg/L、(24.8±4.6)μg/L、(0.54±0.15)%、(10.1±1.3)%比较,差异均有统计学意义(P<0.001)。各指标相关性分析结果显示:ADPey及AA诱导的MPAR、P-Selectin、TXB2、6-k-PGF1a、TP比值与血小板表面CD62P的表达的相关系数分别为0.587、0.672、0.861、0.775、-0.777、0.830(P<0.01)。结论高脂血症促使大鼠血小板活化。除了血小板表面CD62P的表达外,P-Selectin、TP比值能够较好地反映血小板活化的程度。Objective To analyze different platelet activation indexes in hyperlipemia rats, including maximum platelet agglutination rate (MPAR) induced by adenosine diphosphate (ADP) and aracbidonie acid (AA) , P- Seleetin, thromboxane B2 ( TXB2 ) , 6 - keto - prostaglandin F1 a ( 6 - k - PGF1 a), TXB2/6 - k - PGF1 a (TP) ratio and the expression of CD62 on the surface of platelet, and to evaluate the correlation between different indexes. Methods Twenty - four Wistar rats were randomly divided into groups control and high - fat - diet. After eight weeks of intervention, MPAR induced by ADP and AA were detected, the concentrations of P- Selectin, TXB2, 6 -k -PGF1 a were determined by euzymelinked immunosorbent assay, the expression of CD62 on platelet surface by flow eytometrie ( FCM ) analysis. The database was established by Excel software and analyzed by SPSS 11.5 software. Results After high - fat intervention, there was significant difference in ADP - and AA - induced MPAR, P - Selectin, TXB 2, 6 - k - PGFla, TP ratio, platelet CD62P between high - fat - diet group ( 55 ±14%, 30 ±8%, 27.0 ±2.8μg/L, 54 ±9μg/L, 12.6 ±2.8μg/L, 3.25 ±0.61, 25.2 ±5.3 %, respectively) and control (24 ±5%, 17 ±3%, 10.7 ±1.7μg/L, 14 ±4μg/L, 24.8 ±4.6μg/L, 0.54 ±0.15, 10.1 ±1.3, respectively ) ( P 〈0. 001 ). The analysis results of different relevant indicators showed that the correlation coefficients of ADP - and AA - induced MPAR, P - Selectin, TXB 2, 6 - k - PGFla, TP ratio and platelet CD62 expression were 0. 587, 0. 672, 0. 861, 0. 775, - 0. 777, 0. 830, respectively ( P 〈 0. 01 ). Conclusion Hyperlipemia spurs platelet to activation in rats. Except platelct CD62p, P - Selectin and TP ratio can reflect better the platelet activation.
