摘 要:分离延胡索生物碱并测定其对肝癌细胞SMMC-7721的体外细胞毒活性。以30%乙醇为溶媒利用微波法提取延胡索中生物碱类化合物,醇提物经过乙醚、NaOH萃取,大孔树脂、硅胶进一步纯化,同时用化学检识法进行定性鉴别,得到水溶性生物碱、脂溶酚性生物碱和脂溶非酚性生物碱3部分。利用MTT方法研究各部分的细胞毒活性。结果表明延胡索脂溶非酚性生物碱组分肝肿瘤细胞杀伤活性最强,对SMMC-7721的生长抑制活性最高,IC50约为35μg/mL。To investigate the separation methods of alkaloids in Corydalis yanhusuo and determine their cytotoxic activities in a hepatocellular carcinoma cell line (SMMC-7721)in vitro. The alkaloids in Corydalis yanhusuo are extracted with microwave by 30% alcohol, the ethanol elution is leached by aether and sodium hydroxide, then purified through macroporous resin and silica gel column chromatography, qualitative identification is simultaneously performed by chemical methods. Total alkaloids are divided into three parts, water-solubility alkaloids, Phenolated alkaloids and nonphenolic alkaloids, and cytotoxic activities of each part are evaluated by MTT assay. The results show that nonphenolic alkaloids in Corydalis yanhusuo have the highest activity. These components maximally inhibite cell growth of SMMC-7721 and the IC50 value was about 35 μg/mL.
