猪极体的保存研究

摘  要:将从屠宰场采集的猪卵巢卵母细胞进行体外成熟培养和体外受精,得到猪第一极体和第二极体。通过形态学观察和台盼蓝染色,探讨不同保存温度(39℃、4℃、-20℃、-196℃)下猪第一极体和第二极体的存活情况。试验结果表明l随着保存温度的降低,猪第一极体和第二极体的存活率随之升高。第一、二极体在39℃条件下大都在几小时内发生退化;4℃条件下保存40h存活率为70%~80%;-20℃保存7d时存活率达90%以上;而在液氮中保存1个月时,第一极体和第二极体的存活率仍接近90%。Porcine first polar body (PbⅠ) and second polar body (PbⅡ) were obtained from matured and fertilized oocytes by in vitro culture of follicle oocytes in ovary from abattoir. The survival of PbI and PblI under different conservative temperatures (39℃ ,4℃, -20℃ and -196℃ ) was identified by morphology and trypan blue staining,which was very important for the further study of porcine polar bodies. The results showed that there were more and more PbI and PblI survived with the decrease of temperature. At 39 ℃, both PbI and PblI degenerated very fast in a few hours. The survival rate was 70% - 80% when conserved for 40 h at 4 ℃. The survival rate got up to 90% when conserved for 7 d at - 20 ℃. Moreover,almost 90% of PbI and PblI could be kept alive for one month at - 196 ℃.
