摘 要:[目的]观察先暴露于不同时间的汽油废气颗粒的大鼠再次受卵蛋白攻击后其迟发相反应的变化。[方法]雄性wistar大鼠随机分为6组,每组10只。A组给予生理盐水攻击;B组给予卵蛋白攻击。C、D、E、F4个组分别吸入汽油废气颗粒各1、2、3、4周后再用卵蛋白攻击。所有抗原攻击结束1周以后除A组用生理盐水激发30min以外其他组均用卵蛋白激发30min,24h后施行乙酰胆碱激发试验测定其气道阻力,然后比较每只大鼠乙酰胆碱激发后最终气道阻力和基础气道阻力的比值(简称气道阻力比值)。最后观察肺泡灌洗液中嗜酸性粒细胞百分比变化、肺组织中IL-5和IFN-γ的浓度以及肺组织的病理变化(气道壁厚度的变化)。[结果]气道阻力比值在B、C、D、E、F组均较A组增高,E、F组较B组增高,之间差异均有统计学意义(F=305.922,P﹤0.01)。气道阻力比值的增高和汽油废气颗粒吸入的时间有正相关性(r=0.938,P﹤0.01);肺泡灌洗液中嗜酸性粒细胞百分比经统计学分析,B组较A组增高,E、F组较B组增高,之间差异均有统计学意义(F=16.96,P﹤0.01);肺组织中IL-5在D、E、F组较A、B两组增高,差异有统计学意义(F=4.26,P﹤0.05);而肺组织中IFN-γ在D、E、F组较A、B两组降低,差异有统计学意义(F=12.098,P﹤0.05);大鼠气道壁厚度在B、C、D、E、F组较A组增高,E、F组较B组增高,之间差异均有统计学意义(F=106.233,P﹤0.01)。各组大鼠气道壁厚度与吸入时间呈正相关(F=106.233,r=0.894,P﹤0.01)。[结论]DEP吸入后的大鼠再次受卵蛋白攻击后其迟发相反应明显增强,其反应增强程度与DEP吸入时间呈正相关。[Objective]To observe the change of late reaction degree for rats with prior exposed diesel exhaust particulates then re-sensitised by ovalbumin. [Methods] Male Wistar rats were divided into 6 groups randomly. There were 10 rats in each group. A group was sensitised with saline and B group was sensitised with ovalbumin. C, D, E, F groups respectively inhaled DEP for 1 week, 2 weeks, 3 weeks and 4 weeks. After DEP inhaled, rats were re-sensitized with ovalbumin.1 week after antigen sensitizations had done, with exception of A group (A group was stimulated with saline 30 minutes), each group was stimulated with ovalbumin 30 minutes. 24 hours later, airway resistance was measured by acetylcholine provocation test. Compared the ratio of final airway resistance and basic final airway resistance (resistance ratio) of all rats. Finally observed the change of eosinophil in alveolus lavage fluid, concentration of IL-5 and IFN-γin lung tissue and pathologic change (change of airway size). [Results] The resistance ratio of respiratory tract in B, C, D, E, F group was much more increased than in A group, and in E, F group was increased much more than in B group (F=305.922, P﹤0.01). The level of resistance ratio had positive correlation with DEP inhalation time. Eosinophils percent of B group was higher than that of A group. And that of E, F groups were higher than B group. There was significant difference (F=16.96, P﹤0.01); Il-5 concentration of lung tissue was higher in D, E, F groups than in A, B groups, there was significant difference (F=4.26, P﹤0.05); but IFN-γ level was decreased in D, E, F groups than in A and B groups, there was significant difference (F=12.098, P﹤ 0.05). Airway paries thickness percentage of B, C, D, E, F group was higher than A group, which of E, F group was higher than B group (F= 106.233, P﹤0.01).
