恒河猴的血清蛋白质电泳及其与人类、家猪的比较研究

摘  要:[目的]恒河猴(Macaca mulatta)是异种移植研究和药物临床前评价中最重要的动物模型。为了探索临床前研究与临床应用的差距的可能原因,初步研究了恒河猴的血清蛋白质。[方法]分别采集36只正常恒河猴(成都平安动物繁育基地)、30只正常香猪(贵州大学农学院养殖场)和46个正常人(华西医院)的静脉血,采用REP电泳系统电泳血清蛋白。结果采用SPSS 12.0统计学软件分析。[结果]猴血清蛋白中的Alb百分含量比人的低、而比猪的高(P﹤0.05),Alpha1、Alpha2百分含量比人的高、而比猪的低(P﹤0.05),Beta百分含量比猪和人的都低(P﹤0.05),Gamma百分含量比猪的低(P﹤0.05)。[结论]结果显示了猴与人、猪在血清蛋白组分上都存在着一定的差异。这些差异可能会致在猴体内进行的药物临床前研究结果与在人体内的真实情况不符,从而影响药物的临床应用价值和安全性评价;也可能是猪到猴或猪到人的异种移植中生物功能不匹配的潜在原因,而猴和人的差异也可能影响异种移植后的监测和对排斥强度的估计。[Objective]Macaca mulatta (Rhesus monkey) is the most important animal model in the study of the xeno-transplantation and the pre-clinic evaluation of the medicine. To explore the differences between the pre-clinic study and the clinic appliance, we initially studied the quantities and the component of proteins in rhesus serum. [Methods] The venous blood were sampled from 36 healthy rhesus (Chengdu Ping’an Animal Breeding Base), 30 healthy miniature pig (Nursery of Agriculture College in Guizhou University), and 46 healthy human (West China Hospital of Sichuan University), respectively. The serum protein components were separated with Rapid Electrophoretic Analyzer (REP). The results were analyzed by statistic software SPSS 12.0. [Results] The percentage of Alb, one kind of protein components, in rhesus serum was lower than that of human, but higher than that of human (P﹤0.05). The percentages of Alpha 1 and Alpha 2 globulin (Alpha 1, Alpha 2) in rhesus serum were higher than those of human, but lower than that of pig (P﹤0.05). The percentage of Beta globulin (Beta) in rhesus serum was lower than those of pig and human (P﹤0.05). The percentage of Gamma globulin (Gamma) in rhesus serum was lower than that of pig (P﹤0.05). [Conclusion] The results above show the differences between rhesus and human, rhesus and pig in both quantities and components of the serum proteins. On one hand, these differences maybe result in the discrepancy of the drug pre-clinic studies proceeded in rhesus model from the human real situations in pharmacology, metabolism and toxicity, and maybe influence the drug clinic appliance values and the evaluations of the safety. On the other hand, the differences also would be one of the potential causes of the biological mismatch of the xeno-transplantation from pig to monkey or from pig to human, and the differences of rhesus and human would be affect the post-graft monitor after xeno-transplantation and the estimation of the intensity of
