微柱凝胶技术在IgG抗A、抗B效价测定中的应用

摘　　要：目的应用微柱凝胶抗人球蛋白卡(以下简称微柱卡)测定孕妇血清(浆)中的IgG抗A、抗B效价,以预防ABO血型不合引起的新生儿溶血病。方法同时应用微柱卡和凝聚胺法对40份申请IgG抗A、抗B效价测定的孕妇O型血标本进行检测,结果用统计学方法处理。结果两法测得的IgG抗A、抗B效价差异均有显著性(P<0.01),微柱卡法检测IgG抗A、抗B效价的灵敏度分别是凝聚胺法的7.8倍和6.7倍。微柱卡法测得IgG抗A、抗B效价均高于凝聚胺法2～3个梯度。结论微柱卡法可以常规用于IgG抗A、抗B效价测定,且操作简便、快速,结果直观,但由于灵敏度过高,应把正常参考值(64(传统的正常参考值)改为(256。Objective To measure the titer of anti-A and anti-B of IgG in serum of pregnant women by microcolumn gel antiglobulin test cards, and prevent the neonatal hemolytic disease resulted from incompatibility of ABO blood groups. Methods 40 serum samples of pregnant women of O blood type that have been asked for measuring the titers of anti-A and anti-B of IgG were measured with the microcolumn cards and polybrene kits simultaneously. All results were analyzed by statistics method. Results The distinction between two methods for measuring the titers of anti-A and anti-B of IgG were significant difference（ P 〈 0.01 ）, the sensitivities of the microcolumn cards were 7.8 and 6.7 times as high as polybrene test in measuring the titers of anti-A and anti-B of IgG,respectively. The result by microcolumn cards was beyond 2 to 3 gradients than by polybrene kits. Conclusion The microcolumn cards can be used for conventional practice in measuring the titers of anti-A and anti-B of IgG with a advantage of convenient, high-speed and direct viewing in manipulation. The normal consultation number was supposed to be changed from 64 to 256 to balance its sensitivity.
