摘  要:目的:初步观察抗共刺激分子CD137单克隆抗体对分离培养的人外周血CD4+CD 25+T淋巴细胞自噬、凋亡、胀亡现象的影响及其表面特征性标志物FOXp3的表达的变化。方法:采用密度梯度离心法及尼龙棉柱法分离健康志愿者外周血T淋巴细胞,磁性细胞分离器(MACS)分离得到CD4+CD25+T淋巴细胞,分别利用电镜及流式细胞仪观察、检测各分组(抗CD137单克隆抗体2μg/ml组、IgG 1同型抗体对照组)的细胞的自噬率、凋亡率、胀亡率及FOXp3的表达。结果:M ACS可成功分离CD4+CD25+T淋巴细胞,纯度可达80.3%-89.5%;抗CD 137单克隆抗体可促进人外周血CD4+CD25+T淋巴细胞自噬率及凋亡率增加;各组间的胀亡率改变及FOXp3表达差异无统计学意义。结论:抗CD137单克隆抗体可促进CD4+CD25+T淋巴细胞凋亡和自噬,其作用机制未发现与胀亡显著相关,而且不影响CD4+CD25+T细胞中Foxp3的表达。Objective:To observe the autophagy, apoptosis, oncosis rate and their surface expression of FOXp3 in human peripheral blood CD4+CD25+T lymphocytes induced by anti-CD137 monoclonal antibody. Methods . Peripheral blood T lymphocyte of healthy adults were separated by Percoll (1. 073g/ml) and harvested by using nylon column. Then, CD4+CD25+T lymphocytes were isolated by MACS(magnetic cell sorting). The cultured cells were divided into different groups (CD137 mAb group 2 μg /ml, IgG1 isotype control group) and at different time points(0h,72 h) , the autophagy, apoptosis, oncosis and FOXp3 rate were analyzed by electron microscope and flow cytometry. Results: The purity of CD4+CD25+T lymphocytes seperated by MACS was 80.3 % 89.50%. The autophagy and apoptosis rate of peripheral CD4+CD25+ T lymphocytes were increased by CD137 mAb(P〈0. 05). There was no obvious change of oncosis rate and FOXp3 among different groups (P〉 0. 05). Conclusion: Anti-CD137 mAb can increase the autophagy and apoptosis rate of peripheral blood CD4+CD25+ T lymphocytes, but have no effect on Foxp3 expression, and the mechanism does not involve the oncosis.
