摘  要:目的:本研究旨在观察姜黄素对脂多糖(LPS)刺激下的人肾小管近端上皮细胞分泌的单核细胞趋化因子(MCP-1)及白细胞介素8(IL-8)的表达水平变化,并初步探讨其作用机制。方法:将肾小管上皮细胞(HK-2细胞)培养于KSF培养液中,设正常对照组给予正常培养液,LPS刺激组(1ng/ml,100ng/ml和10μg/ml),姜黄素干预组(LPS100ng/ml+姜黄素5μM或50μM),分别培养4~24h后收集细胞,应用Real-time PCR检测各组细胞MCP-1及IL-8的表达。ELISA检测细胞培养上清液中MCP-1及IL-8的表达。结果:在不同浓度的LPS刺激HK-2细胞24h后即可明显升高MCP-1及IL-8的mRNA表达水平,随着LPS浓度(1ng/ml,100ng/ml和10μg/ml)增加,MCP-1 mRNA表达分别上升为对照组的1.74、2.15和14.7倍(P<0.01);IL-8 mRNA表达上升为对照组的2.74、5.4和16.45倍(P<0.01)。而以100ng/ml的LPS刺激HK-2细胞4~24h,观察姜黄素的干预作用,可见不同浓度的姜黄素(5μM和50μM)均可显著抑制LPS所诱导的HK-2细胞MCP-1及IL-8 mRNA的表达,且以50μM姜黄素干预组为明显。同时ELISA检测细胞上清液中MCP-1蛋白水平,可见5μM姜黄素组在24h时较相应对照组下降9.5%(P<0.05),而50μM姜黄素组在8h时即较相应对照组下降19.5%(P<0.01)。而在4h和12h时,5μM的姜黄素干预组较相应对照组IL-8的表达下降的11.72%(P<0.05)和7.58%(P<0.01),在50μM姜黄素干预组则为32.90%和18.79%(P<0.01)。结论:姜黄素可抑制LPS所诱导的肾小管上皮细胞MCP-1及IL-8的表达,提示姜黄素在肾脏炎症过程中具有抑制肾小管上皮细胞炎症因子分泌及潜在抗纤维化作用。Objective:To investigate the effect of curcumin on the expression of monocyte chemoattractant protein-1 (MCP-1) and interleukin 8 (IL-8) in renal tubular epithelial cells induced by lipopolysaccharide (LPS). Methodology:Renal tubular epithelial cells (HK-2 cells) were cultured in KSF medium and pre-treated by LPS with different concentrations (1 ng/ml,100 ng/ml and 10 μg/ml) with or without curcumin (5 μM or 50 μM) for different period. The expression of MCP-1 and IL-8 mRNA was measured by realtime PCR. The level of MCP-1 and IL-8 protein in supernatant was detected by ELISA. Results:The expression of MCP-1 mRNA was significantly increased by 1.60,2.15 and 14.7 times (P〈0.01) after stimulating by LPS (1 ng/ml,100 ng/ml and 10 μg/ml) for 24 hours,respectively. The expression of MCP-1 mRNA was also significantly increased by 2.74,5.40 and 16.5 times (P〈0.01),respectively. By pre-treating with curcumin (5 μM and 50 μM),the up-regulated MCP-1 and IL-8 mRNA expression induced by LPS (.100 ng/ml) could be significantly inhibited,especially in the group with curcumin 50 μM. The expression of MCP-1 in cellular supernatant was decreased 9.50% (P〈0.05) after pre-treating with 5 μM cucurmin for 24 hours,and decreased 19.5% (P〈0.01) after pre-treating with 50 μM cucurmin for 8 hours as comparing with LPS alone. The expression of IL-8 in cellular supernatant was decreased 11.7% (P〈0.05) and 7.58% (P〈0.01) in the group with 5 μM cucurmin,and decreased 32.9% and 18.8% (P〈0.01) in the group with 50 μM cucurmin for 4 hours and 12 hours respectively. Conclusion:Curcumin could partially inhibit the expression of MCP-1 and IL-8 induced by LPS in renal tubular epithelial cell. It suggests that cucurmin could inhibit the secretion of pro-inflammation factors in renal tubular cells during renal inflammation and have a potential anti-fibrosis effect.
