摘 要:目的研究西维美林对大鼠颌下腺和舌下腺放射治疗(放疗)后预防龋齿的作用。方法SD大鼠随机分为5组(n=10):阴性对照组(未放疗、未给药)、单纯给药组(未放疗)、放疗前给药组、放疗后给药组和阳性对照组(放疗+等体积生理盐水)。放疗前1d,大鼠口腔人工接种致龋的茸毛链球菌6715。放疗时,将大鼠颌下腺和舌下腺置于X-射线下,剂量为15Gy,剂量率2~3Gy·min-1,放疗1次;给药条件为:西维美林60mg·kg-1·d-1,ig,tid,于放疗前7d或放疗后当日开始给药。放疗后d28,按Keyes评分法评定大鼠平滑面龋齿和沟龋齿状况,并检测各组大鼠体重的改变情况。结果放疗前或后给予西维美林,大鼠平滑面龋齿和沟龋齿的评分均低于阳性对照组(P<0.05);放疗前给药组评分显著低于放疗后给药组(P<0.05)。放疗前或放疗后给予西美维林对大鼠体重的影响显著小于阳性对照组(P<0.05)。结论西维美林对放疗所致大鼠龋齿有预防作用,放疗前使用比放疗后使用效果好。AIM To study the preventive effect of cevimeline on caries induced by X-ray irradiation to submandibular/sublingual (SM/SL) gland in rats. METHODS Fifty female SD rats were randomly divided into five groups (n = 10) : negative control group (rats without irradiation and drug administration), eevimeline only group (rats without irradiation, but administrated with cevimeline), cevimeline before irradiation group, eevimeline after irradiation group, and positive control group (rats with irradiation and administrated with equal volume of sodium chloride). All rats were infected with Streptococcus sobrinus 6715 one day before irradiation. When irradiated, rats' SM/SL glands were ex to X-ray, the dose was 15 Gy, dose rate was 2 - 3 Gy.min^-1, irradiated for only one time. The dose of cevimeline was 60 mg.kg^-1.d^-1, ig, tid, used seven days before or immediately after irradiation. Twenty-eight days after irradiation, Keyes method was used to evaluate the status of smooth surface caries and pit-and-fissure caries. The changes of rats 'weight were measured. RESULTS Scores of caries in both the cevimeline before irradiation group and the cevimeline after irradiation group were significantly lower than those in the positive control group (P 〈 0.05), while scores of caries in the cevimeline before irradiation group were lower than those in the cevimeline after irradiation group (P 〈 0.05). The changes of rats'weight were smaller in both the cevimeline before irradiation group and the cevimeline after irradiation group than those in the positive control group (P 〈 0.05). CONCLUSION Cevimeline appears to exert its caries-protective effect when pre-treated before X-ray irradiation.
