摘  要:采用PCR-RFLP、CRS-PCR和DNA测序技术,对435头荷斯坦牛的κ-酪蛋白基因的外显子4和5上2个单核苷酸多态性(SNP)位点进行了研究。结果发现,在435头牛群中,共有8种基因型组合。单个SNP位点关联分析表明:T10996A突变位点3种基因型间TA基因型个体乳脂率极显著高于AA基因型个体(P<0.01),TA基因型个体乳蛋白率显著高于AA基因型个体(P<0.05)。T12980C突变位点的3种基因型间TC基因型个体乳脂率极显著高于TT基因型个体(P<0.01),TC和CC基因型个体乳蛋白率显著高于TT基因型个体(P<0.05)。T10996A/T12980C两种基因型组合分析中,在乳脂率上,TTCC基因型组合个体最高,TATC基因型组合个体位居第三位;在乳蛋白率上,TATC基因型组合个体显著高于AATT基因型组合个体(P<0.05)。κ-酪蛋白基因不同基因型和基因型组合与荷斯坦牛乳脂率和乳蛋白率存在相关性,但与产奶量和脂蛋白比不相关。By PCR-RFLP, CRS-PCR and sequencing, the single nucleotide polymorphism (SNP) of two loci in K-casein gene exon 4 and 5 in 435 Holstein cattle were analyzed. The results showed that at locus T10996A, the cow's fat rate with genotype TA was very significantly higher than those with genotypes AA (P 〈 0.01); the cow's protein rate with genotype TA was higher than those with genotypes AA (P 〈 0.05). At locus T12980C, the cow's fat rate with genotype TC was very significantly higher than those with TT genotype (P 〈 0.01); the cow's protein rate with genotypes TC and CC were higher than those with genotype TT (P 〈 0.05). Eight genotype combinations were found in 435 Holstein cattle,the cow's milk fat percentage with genotype combinations TT/CC and TA/TC was the highest and the third, respectively. The cow's milk protein percentage with genotype combinations TA/TC was higher than those with genotypes AA/TT (P〈 0.05). Different genotypes and genotype combinations had correlation with fat rate and protein rate at these two loci, but the polymorphisms had no correlation with milk yield and fat protein ratio.
