穿山龙对胰岛素抵抗糖尿病大鼠血糖改善作用及其机制

摘　　要：目的:观察穿山龙对胰岛素抵抗的糖尿病大鼠的治疗作用。方法:以高脂饲料+小剂量链脲佐菌素(30mg/Kg)制备胰岛素抵抗的糖尿病大鼠模型,随机分为穿山龙组、文迪雅组、对照组。正常组一直以普通饲料饲养。实验组分别给予穿山龙、文迪雅、蒸馏水灌胃,至血糖下降至正常组平均水平或与造模时相比有显著性差异为治疗有效,有效者处死,若至总灌胃时间3个月时则无论是否治疗有效均处死,并测定不同时期的体重、尿素氮、肌酐、总胆固醇、甘油三酯、胰岛素。结果:高脂饮食1月斤实验组大鼠体重及胰岛素明显升高,注射STZ后实验组大鼠血糖明显升高:药物干预后各实验组大鼠各实验阶段GLU、BUN、CRE、TC、TG、ins和体重的差异均有显著性,其中穿山龙组利文迪雅组GLU明显下降,穿山龙纤血-糖较治疗前平均下降8.49mol/L(约40%),文迪雅组血糖下降7.03mmol/L(约33%):两组胰岛素均明显高于对照组,且文迪雅组高于穿山龙组,但均未达到正常水平;各实验组的TC、TG均明显下降。结论:穿山龙与文迪雅有相似的降糖作用,且胰岛素增敏作用可能优于文迪雅。Objective ： Observation of Dioseorea nipponica Makino in the treatment of diabetic rat.Methods：The diabetic rat model was established by high fat diet and injection of streptozotocin.The we randomized divided the diabetic rat into Dioscorea nipponica. Makino group,rosigli.tazone .and control group. Normal group was feeded with common diet.The three experimental groups were given Dioscorea nipponica Makino,rosiklitazone and distilled water respectively.When the rats＇ plasma glucose were decreased to normal level-or there was significant difference when compared to model was defined as effectiveness.As soon as the effectiveness they were killed.If is was three months the rats were given medicine,they would be kiIled whatever it was effective or not.We also measured weight.blood urea nitrogen（BUN）,creat-inine（CRE ）,total cholesterin（TC）,triglyceride（TG）,insulin（Ins） of differentstage of the rats.Results：When the rats＇were feeded with high fat diet after one month,the weight and insulin were increased significantly.After they were injected with streptozotoin,blood g.lucose was increased significandy.After given medicine,the differences of GLU,BUN,CRE,TC,TG,Ins and weight of the experimental .group of different stages of the trial were significanfly.GLU of Dioscorea nipponica Makino_group anft rosiglitazone group were decreased significantly. Dioscoreafnipponica M, akino group decreased 8S49mol/L（about 40%）, and rosiglitazone group-decreased 7.03mmol/L（.about 33%）,Ins of the two groups were significantly higher than control group,Ins of rosiglitazone group was higher than that of Dioscorea nipponica Makino group,but they were all less than normal level.TC .and IG of the expenimental .groups were all- decreased significantly.Conclusions： Dioseorea nipponica Makino has similar effect of reduce blood glucose with rosiglitazone and it＇ s effect of increase the effect of insulin may be prior to that of rosiglitazone.
