摘 要:目的建立HCMV感染Balb/c小鼠椎间盘组织模型,从免疫学、基因表达、组织学等方面来探讨HCMV感染致椎间盘组织变性的可能的机制。方法(1)随机选取6-8周SPF级Balb/c小鼠84只,建立5个不同感染量HCMV AD169株感染Balb/c小鼠模型雌雄各6只为感染组(A组),同时设立病毒灭活组(B组)和细胞对照组(C组)雌雄各6只;(2)间接ELISA法检测小鼠血清中的IgG、IgM抗体水平,取各组小鼠椎间盘组织进行病毒分离,采用PCR和RT-PCR分别检测HCMV UL83基因和UL54基因转录产物;(3)取各组小鼠椎间盘组织行病理学观察。结果A组和B组小鼠IgG抗体均为阳性,C组为阴性,小鼠IgM抗体仅A组中2×109PFU.L-1和2×108PFU.L-1感染组雌性小鼠为阳性,OD值的组间差异有统计学意义(P〈0.05);上述两感染组小鼠病毒分离、PCR及RT-PCR均为阳性,病理学观察示小鼠椎间盘组织出现明显的病理变化;雄性小鼠、其余A组、B组和C组均无明显变化。结论HCMV AD169株能够感染Balb/c小鼠椎间盘,感染与否跟病毒的感染量有关;HCMV活动性感染与椎间盘变性有相关性。 该模型对椎间盘变性的分子水平研究有显著的优势,为椎间盘变性动物模型的建立提供了新的思路和平台。Aim To construct a model in which Balb/c mice are infected with HCMV to explore the possible mechanism in the degeneration of HCMV infecting Balb/c mice intervertebral disc(IVD) tissues from three aspecting of histologic,gene expression and immunology study.Method First,eighty-four Balb/c mice,aged six or eight weeks old,were picked up at random at the level of SPF.Models of 6 females and 6 males by 5 distinct doses of HCMV AD169(group A),inactive virus control group of 6 mice(group B)and cell control group of 6 mice (group B)and cell control ELISA. IVD tissues from each group of mice were picked up to undertake viral isolation and assessment. HCMV' s gene UL83 and UL54 were respectively detected, and corresponding genes' transcription was detected by PCR and RT-PCR. Third, IVD tissues were taken to pathological section with HE stain to make observations. Result IgG in mice' s blood serum of group A and B was masculine, while that of group C was negative;IgM in mice' s blood serum in virus groups of 2 ×10^9 and 2×10^8 PFU · L^-1 was masculine. Difference of OIMS0 value among three groups mouse had statistical significance. In the virus-infected groups above, virus was separated and PCR, RT- PCR were masculine in female mice. The pathological examination showed that there were special changes of pathology in the IVD and that there were no special changes of pathology in male mice, other dose group A,group B and C. Conclusion HCMV can infect mouse IVD and is related to the infected virus dose. IVD degeneration is associated with HCMV infection. This model shows significant advanta- ges for the study of disc degeneration at the molecular level. It provides new ideas and a platform for establishing IVD degenerated model .
