吸入白细胞介素-5对哮喘患者血清可溶性粘附分子-1水平的影响

摘  要:目的:观察白细胞介素-5(IL-5)经雾化吸入后对过敏性哮喘患者和正常人血清中可溶性粘附分子-1(sICAM-1)水平变化的影响。方法:于IL-5吸入前、吸入后2、24、48及72h分别收集8例哮喘患者和6名正常对照者的血清,并以酶联免疫吸附法测定其中sICAM-1水平。结果:IL-5对正常人血清中的sICAM-1水平均无影响。哮喘患者吸入IL-5后2hsICAM-1即明显增高,48h后达最大值,而这种高水平的sICAM-1至少可以维持72h。结论:IL-5可以明显提高哮喘患者而不是正常人的sICAM-1水平;Objective:To observe the effects of inhaled interleukin 5(IL 5) on the changes of concentrations of soluable intracellular adhesion molecule 1 (sICAM 1) in serum from both allergic asthmatics and non allergic healthy subjects. Methods:In a randomized double blind, placebo controlled study design, eight nonsmoking patients with allergic asthma and six normal control subjects were administered recombinant human IL 5 by nebulization, and the concentrations of soluble(s)ICAM 1 in serum from each subject were determined before, and at 2, 24, 48, 72h after inhalation, respectively. Results:sICAM 1 levels in serum within the control group did not change from the baseline at any time throughout the study. In allergic asthmatics, vehicle challenge was not able to cause any changes in sICAM 1 levels in serum. However, there was an increase in serum sICAM 1 levels after IL 5 inhalation, which was significantly greater than the baseline value, reaching a maximum at 48h, and lasted no less than 72h. Conclusions:IL 5 was capable of inducing elevations of serum sICAM 1 concentrations in allergic asthmatics, but not in normal control subjects. The results also suggest that sICAM 1 might he an important effector in the mechanism by which IL 5 induces airway hyperresponsiveness and eosinophilic inflammation in allergic asthmatics.
