摘  要:目的观察原花青素(PC)对缺血再灌注损伤大鼠保护作用及其对中性粒细胞浸润、自由基含量和脑组织含水量的影响。方法将大鼠随机分为假手术组、缺血再灌注组、PC低剂量治疗组及PC高剂量治疗组,线栓法建立局灶性脑缺血再灌注模型。用生化法观察缺血90min再灌注24h大鼠额顶部皮质髓过氧化物酶(MPO)活性、超氧化物歧化酶(SOD)、丙二醛(MDA)含量及脑含水量变化,并进行TTC染色检测脑梗死体积。结果PC治疗组脑梗死体积较缺血再灌注组减小,高、低剂量组之间差异显著;与缺血再灌注组比较,PC治疗组MPO活性、MDA含量显著降低,SOD活性增加,脑组织含水量降低;高、低剂量组之间差异亦显著。结论PC对缺血再灌注脑损伤具有保护作用,其机制可能与减轻缺血再灌注损伤炎症反应,减轻氧化性损伤有关。Objective: To study the effect of procyanidin on neutrophils infiltration and the content of free radical and brain moisture of cerebral ischemia reperfusion (I/R) injury in rats. Methods: The rats were randomly divided into 4 groups: sham operated group, I/R group, low and high dose proeyanidin treatment group. The focal middle cerebral artery occlusion (MCAO) model was made by suture-occluded method. The MPO activity and the content of SOD, MDA and brain moisture were examined after 90min MCAO following 24h of reperfusion. TTC staining was used to calculate the cerebral infarct volume. Results: Compared with I/R group, the MPO activity, the content of MDA and brain moisture of low and high dose proeyanidin treatment group reduced and the content of SOD increased. The cerebral infarction volume of low and high dose procyanidin treatment group was smaller than I/R group. These all showed a dose-dependent relationship. Conclusion: Proeyanidin can alleviate cerebral ischemia reperfusion injure by decreasing the inflammation reaction and free radicals. It plays protective effect on cerebral ischemia injury.
